Milk yield is an important factor,which has limited growth rate of piglets and pork production and can be increased by domperidone in lactating women and gilts.Two experiments were conducted in this study to assess whether would affect milk yield and quality of milk.and growth performance of its offspring.if multiparous sows treated by DOM during late pregnancy and lactation.A total of sixty-four Landrace×Yorkshire multiparous sows(3-5parities.back-fat thickness and body weight were similar)were randomly assigned to two treatments,which are no treatment(NT.247.84 ± 3.08 kg.n=32),Domperidone treated from d 90 of gestation to farrowing(DOM,248.19 ±4.15 kg,n=32).Sows of DOM added 0.4 mg Domperidone per sow per day per kilogram of body weight from d 90 of gestation to farrowing.After farrowing,0 a total of fifty-five Landrace× Yorkshire sows(because of total number borrfwas lesser than six piglets,7 sows didn't be ccounted)from NT(n=28)and DOM(n=27).According to the BW.back fat and litter size of sows on farrowing,assigned the sows from NT randomly to two treatments.which called Control(CON.n=14)and Domperidone treated during lactation(DOL.n=14).At the same time,assigned the sows from DOM randomly to two treatments.which called Domperidone treated during gestation(DOG.n=13)and Domperidone treated during gestation and lactation(DOB.n=14).(?)om d 3 to d 21 of lactation.sows of DOL and DOB added 0.4 mg Domperidone per sow per day per kilogram of body weight.All sows were restrictively fed 2.8 kg/d of a basal gestation diet during late pregnancy.and sows respectively fed 2.0.2.5.3.0,1.5.4.0 kg/d from d I and d 5 of lactation.Water was available at all times.Blood samples were collected on d 90,d 91.d 96.d 100.d 105.d 109 of gestation and farrowing,d 2.d 4.d 7.d 14.d 21 of lactation.Colostrum samples were collected within 12 hours after farrowing.and milk samples were collected at weaning.MG samples were biopsied on d 90.d 110 of gestation and d 2,d 11.d 21 of lactation.The results were as follows1.Effects of Domperidone Supplementation In late Gestational Diet on Reproductive Performance and Mammary Gland Growth of Sows1)Domperidone had no significant effects on growth and reproductive performance of DOM sows.2)Domperidone had a tendency to increase the fat rate of colostrum(P><0.1),and significantly decreased the IgG concentrations(P<0.05).3)Domperidone significantly increased the serum prolactin of sows on d 91 and d 96 of gestation,and significantly increased the mRNA expression of PRLR,Stat5a,Stat5b,LALBA and CSN2 in the MG on d 100 of gestation(P<0.05),while decreased the mRNA expression of FcRn in the MG on d 2 of lactation.2.Effects of Domperidone Supplementation In Lactation Diet on Mammary Gland Growth of Sows and growth performance of offspring.1)To compared with CON,the ADFI of first week,second week and overall lactation of DOL sows had a tendency to increase(P<0.1);the back fat loss and the litter size at weaning of DOG sows had a tendency to decrease(P<0.1)during lactation;the body loss of DOG and DOB sows significantly decreased(P<0.05)during lactation.2)To compared with CON,the weaning average individual weight and the ADG of the first week,second week and the overall lactation of DOL piglets significantly increased(P<0.05);The ADG of DOB piglets had a tendency to increase in the first week of lactation.3)To compared with CON,sows treated by Domperidone during lactation(DOL)can significantly increased milk production ond 11(73.34%.P<0.05)and 21(32.62%,P<0.05)of lactation.4)To compared with CON.all sows from treatments had no significant effect on the protein,fat,lactose and not fat solid ratio of milk;Sows treated by Domperidone during lactation(DOL)had a tendency to decrease IgM concentrations of milk(P<0.1);and sows treated by Domperidone during gestation and lactation(DOB)can significantly decrease IgG concentration of milk(P<0.05).5)To compared with CON,sows treated by Domperidone during lactation(DOL)can significantly increased serum prolactin concentrations on d 4 and d 7 of lactation(P<0.05).;Sows treated by Domperidone during lactation(DOL)can significantly increased the mRNA expression of PRLR.Stat5a,Stat5b and CSN2 in MG(P<0.05)on d 11 of lactation and had a tendency to increase the mRNA expression of LALBA in MG(P<0.1)on d 11 and d 21 of lactation.On d 11 and d 14 of lactation,sows treated by Domperidone during gestation and lactation(DOB)can significantly decrease the mRNA expression of FcRn(P<0.05)in the MG.Conclusions:1.Although DOM could increase serum prolactin levels and promote MG development of multiparous sows at late pregnancy,it is no extra help on MG development during lactation.2.The MG development could be promoted by DOM on lactation,which could enhance milk yield of sows,facilitate growth of offspring.
