| Canine parvovirus disease is an acute,potent,highly contagious infection disease in dog.Because of the high incidence of dogs,especially in puppies,it caused serious harm to the dog culturing in China.At present,the treatment of canine parvovirus disease mainly relys on the virus antibodies,and with the adjuvant therapy.However,antibodies are expensive,therefore,it is needed to find reserve drugs to cure the disease.Attapulgite clay has mucosal protection and strong adsorption capacity,it can absorb intestinal viruses,bacteria and toxins and other harmful substances.Copper and chromium are also closely linked to the growth and development of animals.The three advantages were combined in this experiment,and the attapulgite clay icon-modified with copper and chromium was prepared for the clinical treatment of canine parvovirus disease,in order to evaluate its efficacy,providing a theoretical basis in order to get affordable drug against canine parvovirus disease.At first,the suspected fecal samples infected by parvovirus were collected and tested by CPV rapid detection reagent strips.The results were positive.The fecal samples were inoculated on the F81 monolayer cells,typical CPE in the infected F81 cells could be seen.The isolated virus were identificated by hemagglutination(HA)test and polymerase chain reaction(PCR).The results showed that the fecal samples were demonstrated positive for CPV.CPV also could agglutinate the pig red blood cells,HA titer was 210.It was certified that the virus from samples did be CPV.Attapulgite clay modified with copper and chromium icon in means of liquid icon-exchanged method was studied.Under optimum conditions,the copper icon-modified attapulgite,chromium icon-modified attapulgite,copper firstly and chromium secondly icon-modified attapulgite,and chromium firstly copper secondly icon-modified attapulgite were prepared.The content of copper and chromium ions after digestion was determinated with ICP spectrometer.The result showed that using copper firstly and chromium secondly icon-modified method could absorb more total copper and chromium with attapulgite than other method.The isolated and identificated canine parvovirus was reproduced by the F81 cell,then subcutaneously inoculating in eight 40 to 60-day-old dogs that CPV was negative.Negative control group was not treated,the positive control group only did adjuvant therapy,three experimental groups which were set to low,medium and high doses fed with the attapulgite clay icon-modified with copper and chromium,2 times a day,combined with adjuvant therapy.The results showed that three to four days after inoculated CPV,the dogs began vomiting,mild diarrhea,depression,the CPV was positive.The individuals of positive control group were dead in the fourth and seventh day after inoculation,with most strongly vomiting,diarrhea and other clinical symptoms.The individuals of low-dose group were dead at the seventh day and the ninth day,clinical score was rising but slowly than the positive control group.The individuals of middle-dose group and high-dose group were improving,the clinical scores were decreasing after the first rise with the passage of infection.Pathological anatomy check showed whether the individuals of the positive control group or low-dose group were severely damaged in their small intestine and mesenteric lymph nodes;but the small intestine and mesenteric lymph nodes were normal at the middle-dose group and high-dose group.The results of intestinal tissue biopsy showed that the villus of the dogs from positive control group and the low-dose group were seriously declined and shortened,there were a large number of inflammatory cells infiltrating around,congested and bleeded in submucosal even the muscular.The villus of the middle-dose group and high-dose group were arranged neatly. |
