Zinc finger transcription is a kind of transcription factor with "finger" structure domain that regulates the gene expression.DHHC(Asp-His-His-Cys)is a highly conserved Cysteine Rich Domain that often acts as the element of the DNA binding protein.DHHC-type zinc finger protein genes belong to C2H2-like class.In plants,the understanding of DHHC type zinc finger proteins is limited.In previous study,Zinc finger DHHC domain-containing protein At4g24630 was regard as one of the target protein of bol-miR172 a,which was involved in the pollen development.Therefore,with bioinformatics tools,a comprehensive analysis of the DHHC-type zinc finger protein gene family was performed in Brassica oleracea,including the identification and character of this gene family and analysis of phylogenetic relationship,chromosomal locations and gene structures.Furthermore,the expression divergence of the six-selected DHHC-type zinc finger protein genes in different organsand five flower bud developmental stages(young to mature bud stage)was analyzed with Semi-quantitative RT-PCR and quantitative RT-PCR.Phylogenetic tree and conserved protein domain analyses of 31 DHHC-type zinc finger proteins showed sequence conservation and divergence in this study.According to the detailed analysis of these identified members,they are divided into four groups except Bol032399/Bol015394 and Bol009994/Bol022882.Expression profiling showed that 6-select genes were expressed differentially across disparate organs and differently in different pollen development.This study revealed that Bol032202,Bol015002,Bol009994,Bol035824 and Bol037914 have been expressed intensively in stem and pollen.Expression patterns in flower buds of different developmental stages revealed that,Bol039449,Bol015002,Bol009994 and Bol035824 exhibited significant differential expression and high-expression levels in fertility line.Detailed analysis strongly indicates that these genes are vital to pollen developmentin Brassica oleracea.Moreover,these results provide a genome data resource for pollen developmental genetic research and breeding inBrassica oleracea.
