Brassica oleracea var.acephala is one of the most important ornamental plants in north China.The leaf shape of Brassica oleracea var.acephala is beautiful and elegant,but there are few studies on the leaf shape of kale.Through literature review and our previous research,we obtained three genes related to leaf shape,ALG10,DHHC and LMI1.This study selected kale round leaf material‘honghe’,lobed leaf‘hongkongque’,selection of different growth stage,using real-time fluorescent quantitative PCR technology,analysis crack leaf related genes ALG10、 DHHC、 LMI1 under the different materials and different growth periods,round leaf varieties and crack crack/round leaf variety expression changes in different development stages,the main results of this study are as follows:1.Eight stages were selected,including 12 h seed imbibing stage,70 h seed germination stage,cotyledon stage,one-leaf stage,two-leaf stage,four-leaf stage,18-leaf stage and rosette stage.ALG10 expression level in all stages of ‘honghe’ and‘hongkongque’ increased first,then decreased and then increased.The highest expression was 82.14 times in cotyledon stage,43.71 times in rosette stage,and 0.889 times in control group.2.The expression level of DHHC gene in eight stages between‘honghe’ and ‘hongkongque’shows an overall trend of first rising,then declining and then rising.The expression in cotyledon stage was the highest,which was 76.11 times of the control group,followed by the expression in rosette stage,which was 13.18 times of the control group.The expression in one leaf stage was the lowest,which was the same as the control group.3.The expression level of LMI1 gene in the eight stages of ‘honghe’and ‘hongkongque’ shows an overall trend of increasing first and then decreasing.Among them,the expression in cotyledon stage was the highest,4.66 times as much as that in the control group,and the expression in the imbibed seed stage was the lowest at 12 h,0.69 times as much as that in the control group.We hypothesized that ALG10 gene and DHHC gene might be related to the formation of leaf splitting traits in kale.This study provided the basis for the prediction of candidate genes for the leaf split traits of kale and the regulation mechanism of leaf shape development.
