The traditional composting time of livestock and poultry manure was long,and the loss of volatile nitrogen such as ammonia nitrogen was serious,causing environmental pollution and resulting in the low available of the nutrient content.Focusing on shortening the composting cycle,improving recycling,producing harmless and high nutrient content of organic fertilizer batch aerobic compost reactor with a volume of 75 L and a controlled,heated,ventilated,stirred and ammonia nitrogen recovered capabilities was designed.The test parameters were evaluated and bacterial flora diversity were analyzed by 16 S rDNA high throughput sequencing.Fecal composting process in three important temperature stages were also evaluated for the bacterial flora diversity changes.The main conclusions of this study are:1.Performance of the aerobic compost reactorThe composting time was 44 days.The temperature of the aerobic compost reactor was significantly higher than that of the natural compost temperature,and the temperature of the reactor was higher than 50 ? for about three days,while the natural compost temperature did not change much.The seed germination rate of rapeseed was increased from 31% with the undigested fertilizer to more than 85%with the aerobic fermented compost.The malodor in the process of fermentation was gradually disappeared and no Salmonella species were detected.The composting system and process were very effective in killing bacteria,and remove in insects nuisance.2.Analysis of microbial diversity changes during the composting process1)Through analysis the diversity of bacterial flora in the three key temperature stages during the composting process by combining the 16SrDNAV4 region characteristics amplified by high-throughput sequencing technology,the following conclusions were obtained: First,the effective data for subsequent analysis were found to have the Effective Tags of)63,469,subsequently,the number of Commentson?Taxon Tags?used for the construction of OTUs reached 61381,and the average number of OTUs per sample was 417.All the individual“goods-coverage”were above0.99.2)For the classification of the relative abundance,the top ten bacteria category were Firmicutes,Proteobacteria,Bacteroidetes,Actinobacteria,Fusobacteria,Chloroflexi,Acidobacteria,Tenericutes,Spirochaetes,Nitrospirae,Others;for the outline classification level,the top ten relative abundance of bacterial classes were Bacilli,Gammaproteobacteria,Clostridia,Flavobacteriia,UnidentifiedActinobacteria,Sphingobacteriia,Betaproteobacteria,Erysipelotrichia,Alphaproteobacteria,Fusobacteriia,Others;for mesh classification level,the top ten relative abundance of bacterial mesh included Bacillales,Aeromonadales,Clostridiales,Flavobacteriales,Sphingobacteriales,Lactobacillales Corynebacteriales,Xanthomonadales,Burkholderiales,Micrococcales,Others;on the taxonomic level,the top ten relative abundance of the bacterial families were Bacillaceae,Aeromonadaceae,Flavobacteriaceae,Sphingobacteriaceae,FamilyXI,Planococcaceae,Corynebacteriaceae,Xanthomonadaceae,Alcaligenaceae,Carnobacteriaceae,Others;in case of relative abundance on the classification level,the top ten bacterial genera included Paucisalibacillus,Oceanisphaera,Ulvibacter,Sporosarcina,Corynebacterium1,Olivibacter,Luteimonas,Paenalcaligenes,Erysipelothrix,Sphingobacterium,Others;Each of these levels of classification,and the potential top microbial strains provided theoretical support for the future development of new chicken manure products.
