Eight heat tolerant bacteria(accession numbers:CC1~CC8) were isolated from the chicken manure compost,collected from Chengdu,Chongqing and Emei,using dilution-plate method.Based on results of physiological and biochemical experiments, such as original medium pH,odor reducing and NH3 producing capacity,three strains (CC1,CC3 and CC8) with high efficient odor reducing ability were obtained.16S rRNA gene polymorphism of these three strains was analyzed by PCR-RFLP method,respectively.Among them,16S rRNA of CC3 strain was also selected to be sequenced and constructed the phylogenetic relationship with nine species,such as,B. badius,B.barbaricus,B.circulans,and et al.,based on 16S rRNA sequence.In order to evaluate the efficiency of odor reducing and the capacity of inhibiting the emission of NH3 of the tested strains,indoor compost was hold.To study the effect of three selected heat tolerant strains,two kinds of treatments(inoculation or non-inoculation of these three strains mixture into the chicken manure) were designed, and the fermentation temperature,compost water content,pH value,NH4+-N and organic matter amount were studied during the process of fermentation.The results of the present research were as the following.(1) The RFLP patterns of 16S rDNA fragment digested by MspI among CC1,CC3 and CC8 strains were completely identical.(2) The phylogenetic relationships among nine species,such as B.badius,B.barbaricus,and B.circulans et al.,revealed that the homology of the 16S rDNA sequences between B.badius ATCC 14574T and CC3 was 99.9%.(3) Both the odor reducing efficiency and inhibition of nitrogen losing ability are better than others,when the chicken manure was added as the following ratio:chicken manure 55%,mushroom 30%,coal ash 10%,seedcake 5%).(4) When inoculating CC3 into the chicken manure compost,the volatilizing amount of NH3 was decreased 48.91%compared with the control.However,when inoculating the mixture of three bacteria into the chicken manure compost,i.e.imitating the environment of conventional compost,the amount of NH3 loss was decreased 58.11%, compared with the normal fermentation process.Therefore,these heat tolerant bacteria have potential application prospect for chicken manure fermentation.(5) Compared with CK,the disposal inoculated with the mixed heat tolerant bacteria has not odor,much more white mycelium,lower compost pH value,gaining faster and higher temperature,promoted the water vaporizing;and accelerated the organic matter decomposing during the process of fermentation.Furthermore,the wheat seed germinated 80.1%on the 17th day using these fermentation materials,which showed that the compost reached to the maturity state.The results revealed that inoculation of the mixed heat tolerant bacteria have strong function of deodorization,together with preventing the NH3,and condensation NH4+-N than CK.So the mixed heat tolerant bacteria can be used to chicken manure composting.
