The cupitulum of Coreopsis tinctoria Nutt.have been used as a Uygur medcine and tea in Chinese folklore.The color of tea looks like amber which is rich in pigment.This paper focuses on the polysaccharide from C.tinctoria.1.Phenol-sulfuric acid method were used to determine polysaccharides content and analyse the discrepancy from C.tinctoria and 21 different types of medicinal Chrysanthemum.The results show that polysaccharide contents in C.tinctoria and 21 different types of medicinal Chrysanthemum have significant differences,and no significant difference in different regions C.tinctoria.Scavenging rate effects on OH and DPPH·free radicals of polysaccharides from C.tinctoria compared with 21 types of medicinal Chrysanthemum.It turned out that polysaccharides from C.tinctoria have more powerful scavenging activity than 21 different types of medicinal Chrysanthemum.The antioxidant ability of pH from changji showed that it was slightly weaker than Vc.2.Response surface methodology(RSM)was used to optimize the ultrasonic-assisted extraction technology of polysaccharides from C.tinctoria.Based on single factor experiments,a three-factor-level experiment design were developed by Box-Benhnhen central composite design method with causal factors of ultrasonic temperature,ultrasonic time,ratio of liquid to raw material.The interactive effects of three crucial technological parameters were assessed by response surface methodology(RSM).The optimum ultrasonic-assisted extraction conditions were as follow:ultrasonic temperature was 63?,ultrasonic time was 280 min,the ratio of liquid to raw material was 80:1(mL:g).Under the optimum extraction technology conditions,the extraction value was 2.75 mg-g-1.The conditions are suitable for the extraction process regression analysis and parameter optimization.Response surface methodology(RSM)was used to optimizing the ultrasonic-assisted extraction technology of polysaccharides from C.tinctoria.Based on single factor experiments,a three-factor-level experiment design were developed by Box-Benhnhen central composite design method with causal factors of ultrasonic temperature,ultrasonic time,ratio of liquid to raw material.The interactive effects of three crucial technological parameters were assessed by response surface methodology(RSM).The optimum ultrasonic-assisted extraction conditions were as follow:ultrasonic temperature was 63 ?,ultrasonic time was 280 min,the ratio of liquid to raw material was 80:1 mL-g-1.Under the optimum extraction technology conditions,the extraction value was 2.75 mg-g-1.The conditions are suitable for the extraction process regression analysis and parameter optimization.3.Infrared spectroscopy(IR)and high performance gel chromatography(HPGPC)were applied to explore difference in structure polysaccharides among different regions.HPGPC was used to measure molecular weight from different regions C.tinctoria.The results are as follows:Pishan(Mw=22427),Chang JI(Mw=10832),Turfan(Mw=10090).There are significant difference among them.There exists O-H,and C-H,and C-O,and C-O-H,and sugar ring C-O-C,and beta-Glycoside key structure in polysaccharides with IR.
