For a long time, the kunlun Coreopsis tinctoria by local residents when drink scented tea, has heat-clearing and detoxifying, promoting blood circulation to remove blood stasis, and thefunction of spleen and stomach to treat hot polydipsia, hypertension, flustered, gastrointestinal discomfort, loss of appetite, dysentery and sore furuncle swollen poison, has broad prospects andresearch value.About the Coreopsis tinctoria structure anatomy study has been reported, But about the system ofthe chemical composition of the report has yet to meet. This experiment on the chemical composition of the Coreopsis tinctoria has carried on the preliminary system qualitative experiment research, in order to follow-up on the separation and identification of the chemical composition of the study of pharmacological effects. This study through the Coreopsis tinctoria composition detection, determination of polysaccharide extraction process optimization and activity to the following conclusions:Kunlun Coreopsis tinctoria contains amino acids, protein, carbohydrate, flavonoids, alkaloids, saponins, organic acids, phenols, quinones, volatile oil, grease, steroids, lactones, coumarins components, etc.The single factor tests and response surface optimization method is adopted in the traditional sulfuric acid anthrone colorimetry to determine polysaccharide conditions is studied, it is concludedthat the best determination conditions for:1.1mg/mL anthrone-sulfuric acid reagent addition amount is150μL.The reaction time is36min, reaction temperature96℃. On this condition, the standard curve of the R2is0.9988, the slope is0.0035, intercept is0.01, the linear range is2-26mu g/L, standard addition recovery rate in99.5to100.6%; Different sources of polysaccharide determination results show that the method is of high sensitivity and accurate determination of pol ysaccharide.Water extract-alcohol precipitation are used, ultrasonic assisted extraction and microwave-assisted extraction technology to extract Coreopsis tinctoria polysaccharide, combined with the single factor and response surface method on the extraction process is optimized, it is concluded that thekunlun Coreopsis tinctoria optimum parameters of water extract-alcohol precipitation polysaccharide extraction were as follows:80℃, time of70min and liquid ratio of55ml/g, average totalpolysaccharide extraction rate is8.96%; Optimum parameters of ultrasonic assisted extraction is:the temperature is70℃, time of62min and liquid ratio of55ml/g, ultrasonic power460w, theaverage polysaccharide extraction yield was10.90%; Optimum parameters of microwave assisted extraction for: microwave power is590w, time is8min and liquid ratio of70ml/g, polysaccharide extraction yield of9.95%on average.Polysaccharide in Coreopsis tinctoria extract as the research object, studied the Coreopsis tinctoria polysaccharide removing hydroxyl free radicals, superoxide anion and total antioxidant activity,reducing force and measured the inhibition of alpha glycosidase enzymes activity, The results showed that2.5mg/mL remove hydroxyl free radical activity of polysaccharides in65%,5mg/mLpolysaccharide removal activity of superoxide anion in55%,2.5mg/mL polysaccharide total reducing power at50%,1mg/mL of water extract and methanol, ethyl acetate extract of alpha glycosidase enzyme inhibition activity were74.28%,83.44%and74.92%, respectively.
