Chrysanthemi Flos is the dry capitulum of Chrysanthemum morifolium Ramat,which can be"scattering cold","cleaning heat andtoxin" and "brightening eyes"(.Chrysanthemum mortifolium Ramat cv.)'Hangju' is one of the main culture types of medicinal C.morifolium cv.'Hangju '.The major chemical constituents are flavonoids,volatile oils,chlorogenic acids,etc.It's flavonoids have the ability of scavenging free radicals and superoxide anions,with strong antioxidant activity.Anthocyanins component not only has significant pharmacological activity,but also is the key substanceto study mechanism of flowering.Flood stress is one of the main stress of chrysanthemums growing period often suffered.The past studiesusually focused on flood stress effects onitsseedling stage.Yet there has been no report of medical chrysanthemum'sflood effects in bud differentiation period.The study is about the effects of flood stress to physiology,morphology,and anthocyanin synthesis and related enzymes and genes affect other aspects of C.morifolium cv.'Hangju' cv.'Hangju',with a view to provides a theoretical basis of formation of quality and response to adversity of it.The results are as follows:(1)After the flood stress,the shape of leaves andinflorescence showed no significantchange,while the number of inflorescence remarkably downward withinflorescence developmental malformation.The flood stress also delayed theflorescence of 16 days.Under flood stress,the activity of POD,activity of CAT,and content of MDA increased significantly,and the activity of SOD and content of soluble protein lower than control group.With out floodstress,the activity of POD,activity of CAT,and content of MDA began to decline until returned to control group's value after a period of continuous increase,and activity of SOD and content of soluble protein increased significantly and are higher than control group for some time before returned to control group's value.It shows that flood stress reduced the number of inflorescence and induced inflorescence developmental malformation.Flood stress extened differentiation period of flower bud and delayed anthesis.Under flood press,protective enzyme system,MDA,and soluble protein made a clear response.Afer flood press,the response continued for some time before back to normal levels.The results showed that1320 bp sequence was acquired with the largest open reading frame 1191 bp,which encodes 396 amino acids is CHS of C.morifolium cv.'Hangju' cv.'Hangju'.And 1152 bp sequence was acquired with the largest open reading frame 1029 bp,which encodes 342 amino acids is DFR of C.morifolium cv.'Hangju '.Blast comparison was carried out on NCBI.The genes both had the higher homology compared with the CHSs and DFRs from other species.The expression of CHS in capitulum and different issues of C.morifolium cv.'Hangju'cv.'Hangju' presentthe trend of first rising and after downward with the increase of flowering degree.The expression of DFRin capitulum?ray florets and tubular flower presentthe trend of first downward and after rising with the increase of flowering degree,but in the inflorescence receptacle is reverse.Flood stress Has significant effects on anthocyanins synthesis with relative genes of C.morifoliumcv.'Hangju',but Don't change the patterns of genes expression.The study determinate the content of CHS,activity of DFR and content of anthocyanin.The rusults mean that the content of CHS in capitulum?ray flowers and tubular flowers and inflorescence receptacle present the trend of first rising and after downward with the increase of flowering degree.The activity in capitulum?ray flowers and tubular flowers presentthe trend as follows:downward at first,thenraisedas the flowering degree,but in the inflorescence receptacle is reverse.The content of anthocyanin has the same trend with the activity of DFR.The content of CHS is rising under the flood stress,while the content of anthocyanin and the acyivity of DFR are downward under the flood stress.Flood stress Has significant effects on anthocyanins synthesis with relative enzymeof C.morifoliumcv.'Hangju ',but unchanged the their patterns...
