In this experiment,with the lilium virus-free plants as materials,and the rapid propagation system of lilium had been optimized.The effect of different factors(Phytohormones,Nano-carbon phytohormones,SA,PP333,sucrose and inorganic salt)on differentiation of scales,multiplication and enlargement of bulbs,root formation and physiological characteristics of Lilium were studied.Establishment a high quality and more efficient regeneration system,and provide the foundation for application of Nano-carbon on tissue cultivation.Specific results are as follows:1.With the scales of Lilium as explants,the optimal medium for scales differentiation of Lilium was MS+6-BA 0.5 mg·L-1 +NAA 0.2 mg·L-1 +Nano-carbon 800 mg·L-1 by design test,and the effect of three factors on differentiation of scales was 6-BA>Nano-carbon>NAA.the differentiation rate was 98.09%;2.With the bulbs of Lilium as explants,the optimal medium for bulbs multiplication of Lilium was MS+6-BA 0.8 mg·L-1 +NAA 0.1 mg·L-1 +Nano-carbon 600 mg·L-1 by design test,and the effect of three factors on multiplication of bulbs was 6-BA>Nano-carbon>NAA.the multiplication coefficient was 12.08;3.The effect of different factors on the enlargement of Lilium was sucrose>Nano-carbon>SA>PP333;The best concentration of sucrose for the enlargement of Lilium was 110 g·L-1,bulb fresh weight was up to 1.03 g,was 4.29 times larger than before;diameter was 10.79 mm,and the number of new bulbs was 5.49;4.The optimal rooting media of Lilium was 1/2(macroelements?microelements)MS +0.8 NAA mg·L-1+90 g·L-1 sucrose.The rooting rate was 100%and roots were suitable for transplanting.5.Different concentration of Nano-carbon could all increase the physiological function.The 400 mg·L-1 treatment had the best effect in increasing chlorophyll a,root activity,SOD activity of bulbs and CAT activity of leaves,which were increased by17.4%?29.5%?47.59%and 69.18%;The 600 mg·L-1 treatment had the best effect in increasing dry weight of each parts,chlorophyll b,total nitrogen content of laaves and bulbs,Vc,SOD activity of leaves,CAT activity of bulbs,POD activity of bulbs and leaves,which were increased by19.6%?24.3%?28.2%?24.2%?20.3%?31.89%?53.00%?56.71%?47.67%and 58.72%;The 600 mg·L-1 treatment had the best effect in decreasing MDA content of leaf and bulbs,which were decreased by 49.10%and 37.53%...
