Lilium brownii F.E.Brown Var.viridulum Baker.is perennial herb bulbous plant of Lilium in Liliaceae,also known as musk lily,white lily and medicinal lily.It is one of the three major edible lilies cultivated in China and has important edible,medicinal and ornamental value.In recent years,the planting area of Lilium Longya gradually expanded.Because of the traditional asexual propagation technology,the virus in Lilium gradually accumulated.There are more than 20 kinds of virus diseases that have been reported to infect lily at home and abroad.After infecting lily plants,the virus will lead to the degradation of Lily quality,variety,flower type and color,medicinal,edible and ornamental value of lily,and cause huge economic losses to lily production.In this paper,Lilium Longya was used as the material,the bulb scales of Lilium Longya were used as explants for in vitro culture,and then the optimal culture medium for induction,proliferation and rooting stage of Lilium Longya was selected;the tissue culture seedlings of Lilium Longya were used as the material,the virus-free culture method was used to detoxify the tissue culture seedlings,and the diseases carried by Lilium Longya were treated by molecular biology method Test the poison.The aim is to establish a high efficiency and low cost in vitro regeneration culture system,and lay a foundation for large-scale production and propagation of non-toxic seedlings of Longya Lily.The main research contents and conclusions are as follows:1.In vitro culture experiment of Lilium Longya showed that the upper end of the scales of Longya lily is more suitable for the induction of adventitious buds of lily,The optimal growth medium for adventitious bud induction of lily is MS+1.0 mg/L 6-BA+0.5 mg/L NAA,the differentiation rate is 93%,the differentiation coefficient is 7.43,which is significantly higher than other treatments.Adventitious bud proliferation is greatly affected by exogenous hormones.The best growth medium for adventitious bud proliferation of Longya Lily is MS+1.0 mg/l 6-BA + 0.1 mg/l NAA,and the proliferation coefficient is as high as 6.7.When the concentration of NAA solution is 0.1mg/l,Longya Lily is most suitable for in bottle rooting and out of bottle rooting In this experiment,the best rooting medium was river sand and the best concentration of NAA solution was 300 mg / L.2.Through field investigation,it was found that Lilium Longya in Huaihua city,Hunan province was generally infected by cucumber Mosaic virus,and the symptoms were as follows: yellow and green leaves,severe Mosaic leaves,twisted and deformed leaves,low dwarf plants,etc.,which was consistent with the symptoms after cucumber Mosaic virus infected the plants.Stem tip culture was carried out on lily,and PCR was used to detect the infected plants and stem tip culture plants.About310 bp of specific bands were detected from the infected plants,which was consistent with the expected fragment size.Specific bands were detected in the stem tip culture plants.Therefore,through the experiment of stem tip culture,the length of stem tip was controlled within 0.2-0.5mm,and the cucumber Mosaic virus in the lily plant could be successfully removed.
