With our income increaseing and living standard improvement,the high protein content and low cholesterol content,succulent and tender nutritional richness,delicious taste,easy digestion and good lamb goat of muscle growth is very important.Like other Pax family subfamily,Pax3 and Pax7 in a number of different organisations plays an important role.Pax3 and Pax7 cooperative control of the neural crest,Pax3 is also expressed in the nervous system.Pax3 and Pax7 are the key genes that regulate muscle's development.Pax3 is essential in the formation of early embryos in the process.Whereas Pax7 main regulating postnatal muscle growth and regeneration of injured muscle.Reverse transcription polymerase chain reaction(RT-qPCR)analysis of Pax3 and Pax7 genes in 90 days,180 days and 270 days Tianfu Goat of the semimembranosus,semitendinosus,triceps muscle of arm,biceps femoris muscle,quadriceps femoris,gastrocnemius,the gluteus medius and the deltoid and the longissimus dorsi.The results showed that Pax3 and Pax7 genes were expressed in 9 muscle tissues of Tianfu mutton sheep at different ages.Pax3 has significant tissue specificity.The expression levels of Pax3 mRNA in the forelimb and trunk muscles such as the triceps,longissimus,and deltoid at 90 and 270 days were significantly higher than those hindlimb muscle of the semimembranosus and semitendinosus et.(P<0.05);Pax3 expression was significantly higher in hindlimb muscle of 180 days than in forelimb and trunk muscles(P<0.05).The expression of Pax3 in three periods was also obviously different.The expression of Pax3 in triceps muscle,deltoid muscle and other forelimb muscle decreased at first and then increased.The expression of Pax3 increased first and then decreased in semitendinosus and semitendinosus muscles.The relative expression level of Pax7 in deltoid of three periods was higher.The expression of most muscle tissues in these three periods increased first and then decreased.Reached its maximum at 180 days.At the same time,the expression of MyoD and Myf5 was detected 9 muscle tissues in the above of 90 days,180 days and 270 days Tianfu goat by RT-qPCR technique,and analyze the expression of Pax3,Pax7,Myf5 and MyoD genes in nine muscle tissues.The results showed that the relative expression of Pax7 gene was positively correlated with the relative expression of Pax3,MyoD and Myf5 genes.The relative expression of Pax3 gene was positively correlated with the relativeexpression of Pax7 and Myf5 genes.Western blot analysis showed that Pax3 protein was expressed in nine muscle tissuesat 90 and 270 days.The protein expression of longissimus dorsi,deltoid and triceps brachii was higher than that of thigh,biceps,quadriceps femoris,gastrocnemius,semimembranosus and semitendinosus.However,the protein expression in muscle of 180 day old is opposite.Pax7 protein was expressed in 9 muscle tissues at 90 180 and 270 days.The expression of protein was the highest in triceps muscle at 90 days old.At 180 day old,the expression of longissimus dorsi and semitendinosus muscle was low.The expression of protein in the middle gluteal muscle,triceps muscle,thigh two muscle and thigh fourmuscle of 270 day old was higher than that in other muscle tissuesFrom the results of this study,Pax3 and Pax7 genes play an important role in skeletal muscle growth and development.
