In recent years,the flower industry is growing fast and flourishing gradually in China.As an important ornamental flower plant,the Rosa chinensis jacq attracts a favorable reception because of its unique ornamental value and the effects it contributes to landscaping.Miniature Rose,a special mutant species,which attracts more and more pursuit and attention due to the beautiful pattern and a long flowing gradually.The regulation of the senescence and the shedding of the rose is linked to the potential commercial value of the rose,closely.In this study,we carried out the following studies on RcSAG21 and RcWIN1 genes in petals of rose:1.Two genes related to the senescence were selected from the sequences contained in the database for transcriptome of the Rosaceae,and the genes were cloned from the rose and named RcSAG21 and RcWIN1.RcSAG21 belongs to LEA3 family,which is 420 bp and consist of 95 amino acids furthermore it shows a high homology with wild strawberries and peaches.The product of which is an atypical LEA protein and a stable hydrophobin contains a signal peptide it also may have secretory.The RcWINl gene belongs to AP2 family,which shows a high homology with plum blossom and apple and the length of it was 675 bp and consist of 193 amino acids.The protein analysis shows that the product of the RcWINl gene is an unstable hydrophilic protein,which contains a signal peptide and may have secretory.2.The mechanism of RcSAG21 and RcWIN1 gene in the process of senescence was estimated by analyzing the temporal-spatial expression of the two genes.The expression of RcSAG21 gene was higher in leaves and petals,and increased in three periods of petal.RcWINl gene was expressed in leaves,stems and petals,the expression in petals was higher than the other two parts,and the expression of RcWIN1 was increased rapidly during the fully opened flower stage.RcSAG21 and RcWIN1 genes are the regulatory genes for the senescence of the rose petals and acts on the fully opened flower stage of the petals(during the aging period).3.The effects of exogenous hormones on the senescence of rose were obtained by observing and recording the effects of exogenous hormones on the senescence of the rose,and the mechanism of RcSAG21 and RcWIN1 gene in aging process was supplemented.And the results shows that ethylene can promote the aging of the rose and the connection degree become loose and dropped,moreover the water loss increased.While the effects of 1-MCP were opposite,it can inhibit the aging of the rose and make the connection degree tight,water loss depressed.Exogenous hormones have an impact on the aging process of rose.The mechanism of RcSAG21 and RcWINl gene expression in the process of senescence was further deduced by the analysis of the expression of RcSAG21 and RcWIN1 in the process of budding after stimulated by exogenous hormone.The expression of RcSAG21 gene was improved under the treatment of ethylene and abscisic acid,while the expression of which may be inhibited or show no significant change when treated with 1-MCP and gibberellin.The expression of RcWINl gene was upregulated under the treatment of ethylene and 1-MCP,and no significant change was observed in the treatment of abscisic acid and gibberellin.RcSAG21 gene is a gene that regulates senescence by changing the expression of auxin.Ethylene and abscisic acid can assist in the regulation of auxin to affect senescence and promote the up-regulation of genes.RcWIN1 gene is associated with ethylene signal acceptance,ethylene can promote gene up-regulation,and abscisic acid can't.
