Study On The Function Of FGF11,FGF13 And FGF18 To Hair Follicle Formation And Development In Mouce

According to the classification of hair follicles and mouse hair cycle guidelines,combined with ICR mouse varieties,delimit the test mouse hair follicle induced period at embryonic 13 day(E13),hair follicle start formation at E14,hair follicle formation at E17,hair follicle in anagen at 1-11 day afer birth,12-18 day in catagen,19 and 20 day in telogen,and into next anagen at 21 day.Studies have shown that many members of Fibroblast growth factor family are relate with hair growth and have certain regulat effect in the process of hair follicle development.so far,the role of FGF11 on hair follicle formation and growth has not been reported,FGF13 is expresssed in newborn mice epidermal basal layer and hair follicles,FGF18 is higher expressed at telogen in adult mice skin,but the function on these three genes from hair follicle began to formation to the first hair follicle growth cycle is not system research.So my test choose mouse back skin tissue at E13 to E17,3?5?8?13?16?18?20 and 23 day after birth to detect the localization and expression of FGF11?FGF13 and FGF18 in mouse back skin by immunohistochemistry,RT-PCR and Western bot.The experimental results are as follows:Hair follicle formation:(1)The result of IHC-P showed that the positive reaction of FGF11,FGF13,FGF18 were found in the cytoplasm of basal layer cells,hair germ cells,hari peg,bulb hair follicle cells.FGF13 and FGF18 also expressed in epidermis.(2)RT-PCR?Western blot and IHC-P density statistical data showed that the trend of FGF11,FGF13,FGF18mRNA and their protein were consistent in mouse back skin.The expression of FGF11 was highest at E14,and then the expression was decline.FGF13 present strong staining and highest expressed at E13,then positive reacteion was decline.The expression of FGF18 was lowest at E16,and then the expression was ascend at E17.First hair growth cycle:(1)The result of IHC-P showed that the positive reaction of FGF11,FGF13,FGF18 were found in cell nucleus and cytoplasmic of dermal papilla,hair follicle matrix,inner root sheath,outer root sheath and sweat glands,FGF13 also expressed in hair bulge,basal layer and epidermis,FGF18 also expressed in in hair bulge and epidermis.(2)RT-PCR?Western blot and IHC-P density statistical data showed that the trend of FGF11,FGF13,FGF18mRNA and their protein were consistent in mouse back skin,the expression of FGF11 have not found some regularity.The expression of FGF13 was lowest at 13 day and the expression was ascend from 16 day.The expression of FGF18 was decline from 3 day and lowest at 16 day and the expression was ascend from 18 day.In summary:1 Hair follicle formation:FGF11 play small function at hair follicle induction and play certain role at hair follicle activate;FGF13 not only play role at hair follicle induction,but also regulate the epidermal keratinocytes proliferation;FGF18 may induce hair follicle reaction,and play role at immature hair follicle,promote hair follicles mature.2 First hair growth cycle:postnatal mice skin hair follicle growth cycle need FGF11,but it may have no specific role;FGF13 may have certain effect on hair follicles self-renewal,induct Hair follicles from telogen to anagen stage,promote hair follicle growth;FGF18 plays certain role in hair follicle transition from tolegen to anagen,stimulates the hair follicles into growth phase from quiescence.
