As a new type of antibiotic substitute,antibacterial peptide has the functions of broad-spectrum sterilization and enhancement of animal immunity.It has been widely used as feed additive for animals such as pigs and chickens.This study investigated the effects of the different doses of compound antimicrobial peptide "taikanglibao" on rumen bacterial diversity,microbial community structure and dominant functional bacteria of chuanzhong black goat using MiSeq sequencing technology and Real-time PCR.To investigate the composite antibacterial peptide "taikanglibao" on goat rumen bacterial diversity,community structure and main function of the number of dominant functional bacteria,then provided a theoretical basis for the ruminant feeding that the compound antimicrobial peptides"taikanglibao"as feed additives in ruminant animal.Eighteen male goats[4-month-old,?15.53±1.06?kg]were randomly divided into 3 groups according to the similar weight.Three groups were basaldiet group I(concentrate 300g·d-1·goat-1),group ?(concentrate 300 g·d-1·goat-1+ AMP 3g d-1 goat-1),group ?(concentrate 300 g·d-1·goat-1 + AMP 3 g·d-1·goat-1),continuous feeding of 60 d.The rumen fluid samples were collected after 20 d and 60 d,samples of group I,II and III were labeled as sample A1?A2?A3,sample B1?B2?B3 and sample C1?C2?C3 at 20 d,the samples were labeled as F1?F2?F3,sample G1?G2?G3 and sample H1?H2?H3 at 60 d.And then the total DNA were extraced for amplification 16S rRNA?520F-802R?,sequencing by MiSeq Illumina250,and the number of bacteria in different groups was compared by Real-time PCR.The results showed that:?1?The composite antimicrobial peptidecould "taikanglibao" increase the diversity of bacteria in the rumen of chuanzhong black goat,and the Chao 1 index and ACE index of H were significantly higher than that of F?P<0.05?.?2?Bacteroidetes?40.87%?52.77%?was the most abundant phyla in all samples?except sample H?,followed by the Firmicutes?18.05%?30.39%?and Proteobacteria?except sample H??3.29%?19.99%?;At genus level,Prevotella was the most abundant genus in all samples?25.54%-32.97%?.?3?At phylum level,Firmicutes?Verrucomicrobia and Tenericuteswere significantly increased when supplementation of AMP?P<0.05?,but no significantly difference between two antimicrobial peptidegroups.Verrucomicrobia were significantly higher in H compared with G group?P<0.05?,Proteobacteria were significantly decreased when supplementation of AMP?P<0.05?,and significantly lower in H compared with G?P<0.05?at 60d,At genus level,Prevotella?Butyrivibrio?Succiniclasticumwere significantly increased when supplementation of AMP?P<0.05?,and Butyrivibrio was significantly increased in B compared with C?P<0.05?,while Succiniclasticum was significantly decreased?P<0.05?.?4?Fibrobacter succinogene were significantly increased when supplementation of AMP,and it more obvious in sample H at 60d.Only at 20d,the Ruminococcus flavefaciens and Ruminobacter amylophilus in sample C were significantly increased when supplementation of AMP,and significantly higher in C compared with B?P<0.05?,but no significant difference among three groups.During the whole experimental period,the Ruminococcus albus and Anaerovibrio lipolytica were significantly increased when supplementation of AMP?P<0.05?.Only in 60d,Methanogens and Butyrivibrio fibrisolvens were significantly increased when supplementation of AMP?P<0.05?,but Methanogens in samples H more obviously,Butyrivibrio fibrisolvens in sample G more obviously.Conclusion:Bacteroidetes was the most abundant phyla in chuanzhong black goat,and Prevot ella was the most abundant genus.Composite antimicrobial peptides"taikanglibao" can increase the relative content of Firmicutes?Verrucomicrobia?Tenericutes?Prevotella?Butyrivibrio?Succiniclasticum?Ruminococcus flavefaciens?Ruminobacter amylophilus?R uminococcus albus?Anaerovibrio lipolytica?Methanogens?Butyrivibrio fibrisolvens.This shows that the composite antibacterial peptide"taikanglibao" can regulate the ruminal b acterial community structure,promote some fiber degrading bacteria,fat decomposing bacteria,protein and starch degradation in rumen bacteria growth,enhanced plant fiber,fat,starch and protein digestion,and then improve the growth of the goat.
