Phthalic acid estersor or phthalate esters?PAEs?,also known as phthalates,are the plasticizers produced and used widely in the world.Dibutyl phthalate?DBP?is a common specie of synthesized PAEs.Because of the low cost and high production,it is used in a wide range.With the mass production and using of plastic products,the plastic waste is increasing daily.So DBP follows later.In recent years,it has been found that the exposure of DBP,which is considered an adjuvant,reduces the immune tolerance of the lungs and easily induces allergic airway inflammation.Studies have shown that one of the main mechanisms of the toxic effects of DBP is oxidative stress.When the internal environment of the lungs is disturbed,the immune regulation function of the lungs will be weakened,and it is easy to cause allergic airway inflammation.Therefore,it is important clinical significance that exploring the underlying mechanism of allergic airway inflammation induced by DBP in the lung and targeting the DBP adjuvant on target.Resveratrol?Res?,named?E?-3,5,4-trihydroxystilbene in chemical,is a polyphenolic compound.Res is a phytoalexin produced by itself to resist stress,anoxia,mechanical damage,ultraviolet radiation,and fungal infection during growth.Extensive studies have shown that Res has a biological effect against inflammation,oxidative stress,apoptosis,mitochondrial dysfunction,and angiogenesis.Can Res reduce DBP-induced allergic airway inflammation?The present study was conducted in two ways.?1?The dose-dependent test of DBP?healthy rats,singly sensitized and challenged by ovalbumin?OVA?,combined with low,medium,high doses of DBP and OVA,n=12?.Oxidative stress,airway inflammation,and cell damage were detected in rats exposed to DBP.Wistar rats were intragastrically fed in DBP at 0,500,750,and 1000 mg/kg/day for 9 consecutive weeks,and1 mL/OVA sensitized mixture?1 mg OVA and 100 mg of Al?OH?3 was dissolved in 1 mL of saline?was intraperitoneally injected respectively on the 42nd,49th,and 56th day.At the same time atomization of 1%OVA was applied from day 57 to day 63,30/min/day.The results revealed that compared with the OVA group solely,the one of DBP exposure produces more inflammatory cells?a marked increasement in the number of white blood cells,lymphocytes,neutrophils,and eosinophils?,so does lactate dehydrogenase activity in the serum.Rats exposed in DBP showed more severe levels of oxidative stress,higher concentrations of lipid peroxidation product malondialdehyde,lower concentrations of glutathione,and lower activity of superoxide dismutase.Meanwhile,a Th2-type immune response was induced in DBP rats,and typical Th2 cytokines IL-4and IL-13 were significantly increased,accompanied by an increase in OVA-specific IgE levels.DBP induced a Th2-type immune response in rats.With the levels of OVA-specific IgE increasing,the typical Th2 cytokines IL-4 and IL-13 go up significantly?2?The intervention test of Res?normal and healthy rats,sensitized and challenged jointly by DBP and OVA,treated by Res and Res+DBP+OVA,n=12?.To assess the release function of Res on DBP-induced allergic airway inflammation,intervention experiments that Res?100 mg/kg/day?was given intragastricly to rats exposed to DBP?750 mg/kg/day?with obvious symptoms of the disease are carried out.The results showed that the intervention of Res effectively reduced the number of inflammatory cells in the blood,so do the oxidative injury and Th2-type immune responses of rats induced by DBP?P<0.05?.Res promotes nuclear entry of Nrf2,inhibits the expression of Thymic stromal lymphopoietin?TSLP?and reduces the phosphorylation of STAT6.This in turn reduces the overdifferentiation of Th2 cells and maintains Th1/Th2 balance,thereby alleviating the adjuvant effect of DBP on allergic airway inflammation.In summary,Res activates the Nrf2 signaling pathway.It enhances the body's antioxidant defense system's ability,removes excess active oxygen,reduces the production of TSLP,balances the Th2-type immune response,and reduces DBP-induced rats'allergic airway inflammation.This relieves the pulmonary toxicity of DBP.This study provided a practical basis for the application of Res on the prevention and treatment of allergic airway inflammation in livestock and poultry induced by DBP,which ensures the healthy breeding of livestock and poultry.
