Larix principis-rupprechtii is an important tree species for economic ecological forests in North China.It has the characteristics of fast growth,good material quality and wide use.The Larix seed orchard was created from 60 s to 70 s in China,which provides a large number of germplasm resources for the promotion and study of its genetic diversity.However,the seed production has a long existence of low seed yield and low seed yield.The related research has been carried out.In this paper,296 clones in the seed orchard of breeding base of Larix principis-rupprechti were used as experimental materials,and the method of cone number investigation was used to screen the fine traits of the female clones.The hormone analysis test was carried out with the male and female clones as the control.The development of the male and female flowers was observed and the key period was selected for the transcriptional group.The primers were designed by the sequencing results,and the primers were screened with polymorphism as the index.The main results were as follows:1.The seed setting of 296 clones in Larix principis-rupprechtii seed production showed that there were significant differences in the size of the seed set between different clones,The male clones were 81、125,132,143,149,151,157,169,174,177,194,200,292,294,and the female were 5、20、38、40、57、75、105、110、112、120、209、230、240、247、283.And the yield of the female clones accounted for 70.4% of the total amount.2.The female and male flower buds were photographed with phenotypic photography and paraffin section.A number of clear photographs were obtained.From the external morphological changes and internal anatomical observations,two aspects of the flower bud differentiation and the development of the flower bud were studied,and 3 critical periods were selected for the sequencing of the transcriptional group.3.The hormone determination of female clones No.10 and male clones No.6were selected.It was found that the hormone content of IAA and ABA was significantly different in different periods.The IAA-leucine binding state was very significant in different periods.In other cases,the difference was not significant.4.The male and female flower buds of the male and female clones were sequenced in one key period.Gene expression,differential gene expression and differential gene enrichment were analyzed,and 588 pairs of Larix primers were designed,and 40 pairs of polymorphic primers were screened by EST-SSR molecular marker method. |
