Effects Of Scutellaria (Scutellaria Radix),Pueraria (Puerariae Radix),Isatis Root (Isatidis Radix) And Its Compound On Growth,Repairing Of Oxidized Soybean Oil Injury And CyHV-2 Immunoprotection Of Gibel Carp (Carassius Auratus Gibelio)

In this study,the effects of adding scutellaria(Scutellaria radix),pueraria(Puerariae radix),isatis root(Isatidis radix)and its compound on the growth performance,antioxidant capacity,immunity,oxygen-carrying capacity,coagulation ability,bile acid intestinal-liver circulation,liver and intestinal structure of gibel carp(Carassius auratus gibelio)were studied under the condition of normal or oxidized oil diet.After breeding,we conduct a CyHV-2(Cyprinid Herpesvirus 2)intraperitoneal injection challenge test on gibel carp.This paper is divided into four experimental parts:First part: We add 0.1% of scutellaria,pueraria,isatis root and 0.05% compound natural plant to normal diet,prepare into five kinds of experimental feed: control group(Referred to as “S”),scutellaria group(Scu),pueraria group(Pue),isatis root group(Isa)and compound natural plant group(Cnp).After feeding gibel carp in the pond for 70 days,we analyse the growth performance,antioxidant ability,immunity,oxygen-carrying capacity,coagulation ability and bile acid intestinal-liver circulation,liver and intestinal structure of gibel carp.The results show that,(1).The growth performance of single natural plants shows down trend,while the compound had no significant effect on the growth performance;(2).Natural plants can stimulate antioxidant function of fish.(3).Scutellaria has a prominent role in promoting immune cell proliferation,and the compound can enhance specific immunity of fish.(4).Natural plants can enhance oxygen-carrying capacity of gibel carp,three kinds of single natural plant will reduce the number of thrombocytes in the blood,while the compound will increased the number of thrombocytes in the blood;(5).Three kinds of single natural plant are likely to cause bile acid enterohepatic circulation disorders,leading to cholestasis,while the compound will not have an impact on bile acid enterohepatic circulation.(6).Pueraria and isatis root can cause liver,intestinal injury(Scutellaria cause liver injury slightly),while the compound has no damage to liver or intestinal.(7).0.1% single natural plant cause damage to gibel carp,while the 0.05% compound has no significant effect on gibel carp,suggesting that compatibility and dose of natural plants are closely related to their efficacy.Second part: Gibel carp which breed in first part are intraperitoneally injected with CyHV-2 for 7 days,and samples at 0 d,3 d and 7 d.The changes of immune protection rate,antioxidant ability,immunity,oxygen-carrying capacity,coagulation ability,serum bile acid and liver permeability after attack are analyzed.The results show that,(1).After CyHV-2 infection,gibel carp shows oxidative stress injury,immune damage,oxygen-carrying and coagulation capacity abnormalities,bile acid deficiency and larger liver permeability,and lead to death ultimately.(2).Scutellaria can stimulate the increase of immune cells,maintain coagulation function,improve bile acid deficiency and reduce liver permeability,the relative immune protection rate is 33.3%.(3).Pueraria has a certain role in maintaining coagulation function,improving bile acid deficiency,but it can reduce immune globulin and lysozyme production,increase liver permeability,the relative immune protection rate is-22.2%.(4).Isatis root has a certain effect on stimulating proliferation of immune cells,maintaining coagulation function and improving bile acid deficiency,while the permeability of liver is increased.The relative immune protection rate is 0%.(5).Compound natural plant can reduce oxidative stress damage,stimulate proliferation of immune cells,promote production of lysozyme and reduce permeability of liver.The relative immune protection rate is 66.7%.Third part: Oxidized-basal diet which replace 4% soybean oil with equivalent oxidized soybean oil has been prepared,and then we add 0.1% of scutellaria,pueraria,isatis root and 0.05% compound natural plant to oxidized-basal diet,make into six kinds of experimental feed: soybean oil group(basic feed,referred to as “S”),oxidized soybean oil group(oxidized-basal feed,“OS”),oxidized-basal feed + scutellaria group(OS+Scu),oxidized-basal feed + pueraria group(OS+Pue),oxidized-basal feed + isatis root group(OS+Isa)and oxidized-basal feed + compound natural plant group(OS+Cnp).After feeding gibel carp in the pond for 70 days,we analyse growth and physiological indexes of gibel carp.The results show that,(1).Oxidized soybean oil will lead to growth performance significant decrease,which relate to gibel carp produces oxidative stress,inflammation,decreasing of oxygen-carrying and coagulation ability,cholestasis,liver and intestinal damage.(2).Scutellaria can reduce oxidative stress and inflammatory injury,enhance oxygen-carrying capacity,maintain liver and intestine structure,and increase growth performance to the level of S group.(3).Pueraria has a certain improvement effect on oxidative stress,inflammatory response and oxygen-carrying capacity decline caused by oxidized soybean oil,but the growth performance is still at the same level as OS group.(4).Isatis root can reduce oxidative stress and inflammatory injury,enhance oxygen-carrying capacity,improve cholestasis,maintenance liver,intestinal structure,and increase growth performance to the level of S group.(5).Compound natural plant has a certain improvement effect on oxidative stress,inflammatory response,oxygen-carrying and coagulation capacity decline,liver and intestinal injury caused by oxidized soybean oil,but the growth performance is still at the same level as OS group.Fourth part: Gibel carp which breed in third part are intraperitoneally injected with CyHV-2 for 7 days,and samples at 0 d,3 d and 7 d.The changes of immune protection rate,antioxidant ability,immunity,oxygen-carrying capacity,coagulation ability,serum bile acid and liver permeability after attack are analyzed.The results show that,(1).Compared with S group,OS group shows oxidative stress increase,immunity decline,oxygen-carrying and coagulation dysfunction,liver-damage aggravate,indicating that oxidative oil will increase the body's physiological function damage,which make relative immune protection rate decrease by 11%.(2).Scutellaria can reduce oxidative stress,enhance immunity,maintain oxygen-carrying function,improve bile acid secretion,reduce liver permeability,the relative immune protection rate is 11.1%.(3).Pueraria can enhance immunity,maintain coagulation function,reduce liver permeability,the relative immune protection rate is 66.7%.(4).Isatis root can reduce oxidative stress,enhance immunity,maintain oxygen-carrying and coagulation function,reduce liver permeability,the relative immune protection rate is 55.5%.(5).Compound natural plant can maintain specific immunity,reduce liver permeability,the relative immune protection rate is 77.8%.
