| Pepper is an important economic crop that has been planted all over the world.There are many types of pepper,varying from fruit shape to fruit color,capsaicin content is also different.Pepper is rich in nutrients essential for humans,such as soluble sugar,Vc,capsaicin,and capsanthin.The sequencing of the whole genome of pepper laid a foundation for studying the molecular mechanism of fruit development and fruit color.In this paper,the transcriptome sequencing technology was used to analyze the differentially expressed structural genes of capsaicin and capsanthin pathways in Vc,and the key genes regulating these synthetic pathways were identified through co-expression analysis.The key genes were verified by quantitative RT-PCR.The amount of expression in different organizations at different times.Obtain the following research results:1 RNA-Seq sequencing technology was used to sequence the roots,stems,leaves,flowers,flesh,placenta and seeds of the pepper parent “6421” at different developmental stages.Each sample was repeated 3 times,and more than 150 cumulative sequencing samples were obtained.A total of 1000 Gb Clean reads were obtained,and a large database of pepper RNA dynamic transcriptome visualization database was constructed through bioinformatics analysis."6421" and Zunla’s Alignment are almost all above 90%.The Alignment of CM334 is relatively low,basically between 80% and 85%.2 Comparative analysis of pepper and tomato synthetic Vc pathway structural gene expression in different developmental stages of fruit flesh,pepper fruit Vc synthesis is mainly completed before completely red ripe;GME1 and AO genes play an important role in the Vc content of pepper flesh.3 51 genes involved in the synthesis of capsaicin were analyzed.AT3 d and KasId were specifically expressed in the placenta of capsicum and played a decisive role in the synthesis pathway of capsaicin.Specific gene expression and co-expression analysis suggested that Capana06g001834 and Capana03g000296(bHLH)may be involved in the regulation of capsaicin Synthesis.4 The differential expression of the structural genes of the capsaicin synthesis pathway was analyzed.The LCYB gene was expressed in different tissues of pepper,and the expression levels were not changed at different developmental stages.It was speculated that it not only participates in the synthesis of capsanthin,but also regulates the development of pepper fruits;CCS Genes are specifically expressed in pepper flesh and play a decisive role in the synthetic pathway of capsanthin. |
