C2H2 type zinc finger protein is one of the most classic and extensively studied zinc finger proteins in zinc finger family.The zinc finger protein has a specific motif QALGGH in plants,which indicates that C2H2 type zinc finger protein plays an important role in plant growth and development.Besides,it participates in abiotic stress responses and biotic stress.However,there are only few study about C2H2 type zinc finger protein in the genome of tomato species and the response to drought,salt and cold stress.In this study,bioinformatics methods and software were used to identify the members of the C2H2 type zinc finger family in tomato genome,and a systematic bioinformatics analysis was carried out.At the same time,the expression patterns of some genes were analyzed under tissue specificity and drought,salt and cold stress,which laid a foundation for the further study of C2H2 type zinc finger family.The results showed that:1.99 C2H2-type zinc finger protein genes were identified and classified into four groups based on a phylogenetic analysis.Some physical parameters such as molecular mass,pI value and protein length were calculated for each C2H2-type zinc finger protein.The distribution of introns and 15 motifs in the genome of the tomato C2H2-type zinc finger transcription factors were calculated.Ninety-six of all 99 tomato C2H2-type zinc finger genes were unevenly distributed across all 12 chromosomes.Many abiotic stress-related,phytohormone-related and tissue-specific expression-related cis-elements were analyzed.2.Tissue-specific Expression pattern analysis of 21 typical C2H2-type zinc finger genes showed that SlZF-24 and SlZF-28 were significantly highly expressed in root and that SlZF-6 and SlZF-13 were preferentially expressed in flower.An analysis of responses to abiotic stresses showed that the expression levels of many C2H2-ZFP genes were related to treatment with at least one of multiple abiotic stresses.SlZF-6 and SlZF-67 were induced under all three stresses,and SlZF-20 was suppressed under all three stresses.Fifteen of 21 genes were clearly upregulated or downregulated under these three stresses.3.Building pTRV-SlZF-36 VIGS vector and transducting to tomato seedlings successful,and the qRT-PCR showed that SlZF-36 gene participated in the response process of drought.
