Cucumber?Cucumis sativus L.;2n=2x=14?belongs to cucurbit cucumbers and it is one of the important vegetables in the world.At the same time,it is also one of the important greenhouse vegetable crops in China.The vegetative organs of cucumber,including roots,stems and leaves,although they are not the final edible organs,they have great significance to the final production of the whole plant.The cucumber stem is the middle part of the plant,which has the function of transporting nutrients and moisture as well as supporting leaves,flowers and fruits in the certain space.Cucumber clustered?multiple lateral branching?is a rational plant type in the growth process of cucumber,which is of great significance for the improvement of yield,but relatively few reports have been reported now.The experimental materials in this study were derived from the cluster mutant 430 in the EMS mutant library of the research group.The identification and preliminary localization of candidate gene of cucumber clustered mutant 430 were performed,and the cucumber cluster candidate gene?tentatively named Cs-clus?was initially locatedThe main results are as follows:?1?Observation of the entire growth period of the cucumber cluster mutant 430 revealed that there was a mutation in the plant type and flower organ compared to the wild type.The clustered mutant 430stem was thicker than the wild type,and the stem was polyhedral.There were 2-4 leaves on the same node.Dwarfing was also observed.The wild-type plant height was 66.76±2.58 cm,and the mutant plant height was 56.12±2.69 cm;wild-type male and female petals are all 5,ovary is oblong,and 430 petals are 10,and the ovary is oval.?2?The genetic analysis was performed on the clustering traits:the 430 mutant which crossed with the wild type,The experimrnt showed that F1 represents the normal wild phenotype.Observed the F2generations obtained through self-pollination of the F1 generation,51 were 430 mutant phenotypes,139were wild-type,the segregation ratio was close to 3:1,and the normal plants were detected in the F2generation by Karman??2?.The ratio to the mutant plants corresponds to the segregation ratio of the genetics of Mendel.It was shown that the 430 mutant is a quality trait controlled by a single stealth gene.?3?MutMap method was used for re-sequencing in mixed pools.Which with 30 F2 clustered phenotypes were selected and mixed with equal amounts of DNA to construct a gene pool.Simultaneous re-sequencing with the original wild-type material 649 was performed based on the SNP index and the mutation site pair gene.The impact of the comprehensive analysis of the screening of the cucumber chromosome 1 with 2 SNP is located in the exon region of the C-T mutation point,gene Csa1G059210,Csa1G064750.The KASP genotyping technique was performed in the F2 generation population.It was found that the genes Csa1G059210 and Csa1G064750 were not identical.This shows that the genes Csa1G059210 and Csa1G064750 are not Cs-clus genes.Other candidate genes are currently being identified.
