Sesame(Sesamum indicum,2n=26),which contains abundant nutrition,is one of the most ancient oil crops all over the world.It is significantly valuable in multiple aspects,such as food,medicine,horticulture,health care products,animal husbandry.As of today,8115 pieces of sesame germplasm resources have been collected in China.However,as the standard molecular identification method of sesame and the platform sharing the information of sesame germplasm resource are not available and accessible easily,the sesame germplasm resources couldn’t be utilized efficiently.Therefore,the sesame applied core collection has been established so as to protect and utilize the sesame germplasm resources at high efficiency.Based on the materials from the sesame applied core collection,the method of building DNA molecular identification has been explored,and the information database of sesame germplasm resources has been built simultaneously.That could not only faciliate the efficiency and precision of providing the breeding materials,but also provide full supports in sharing the information of these sesame resources used in the research progress.And that is shockingly meaningful for the fundamental research and breeding improvement.Following are the main results:1.In order to achieve the target of breeding premium varieties,131 pieces of sesame core collection 131 sesame accessions,grouped into 7 different classes,which were high yield,high quality,stress and disease resistant,suitable growth period,mechanization adaptive,specifics of plant morphology,excellent properties overall,respectively,were screened out according to the observation of the sesame core collection grown in 4 various environments for 4 years.2.Based on 32 pairs of SSR primers developed from the sesame genome,7 pairs of primers,bearing high polymorphism,distinct bands and excellent repetition,were screened out using the technique of denatured polyacrylamide gel electrophoresis for detecting SSR and utilized as primary primers so as to precisely identify the applied core collection and explore the establishment of sesame DNA identification.The more precise detection technique of capillary electrophoresis with fluorescent SSR markers was utilized simultaneously in order to analyze the 131 sesame germplasm from applied core collection.Totally 53 polymorphic locations were detected out from 6 primer pairs,and the most sensitive primer pair could detect 12 polymorphic locations.3.The results from the detection of capillary electrophoresis were shown in the form of numbers combined with English letters.As per the rule that the least primer pairs could detect the most sesame germplasm,the combinations of the 6 core primer pairs,namely ZMM1494、ZMM1648、ZMM3037、ZMM2818、ZMM1851、ZMM1935 that could effectively distinguish the 131 sesame germplasm from applied core collection was screened out by ID Analysis 4.0.Following that,the simple and easy-applied DNA molecular identifications such as “4A32645(AC017)” were constructed,and the various combinations of different primers that could distinguish the germplasm within groups were screened out simultaneously.The DNA molecular identifications of character strings,Bar code and Quick Response(QR)codes,easily scanned and recognized by electric gadget,were constructed based on the molecular data of 131 sesame applied core collection so as to broaden their application ranges.They could also provide important technical foundation for the standardization of sesame germplasm and facilitate the construction of DNA identification bank of sesame varieties.4.The online information database of sesame germplasm resources,namely http://www.sesame-bioinfo.org/phenotype/index.html.,where the information of sesame germplasm resources could be browsed and retrieved,the physical sesame resources could be inquired conveniently,was constructed based on lamp database technology combined with the technique of HTML and Java Script.Totally,8115 sesame germplasm resources were registered in the database,including 90420 pieces of information involving fundamental characters,morphologics,biological properties of sesame resources.Besides that,the information concerning sesame core germplasm,micro-core germplasm and applied core germplasm were collected widely and edited as per the strict standard,and the other information on sesame germplasm resources,such as figures,dissertation,thesis,breeding trend and patents were also embedded in the database.The standard,high-precision information involving sesame was available conveniently and physical materials could be inquired by the professional researchers.Simultaneously,it could also be utilized as a platform to facilitate the dissertation of popular science of sesame.
