As an important aquaculture animal,Litopenaeus vannamei cultivation has brought great economic value and social benefits.However,diseases caused by various pathogens have been restricting the healthy development of shrimp aquaculture.Of these,White spot syndrome virus(WSSV),the most serious pathogens of shrimp disease,has been causing cultured shrimp death on a staggering scale since it breaked out.And there is no feasible method to control the occurrence and spread of this virus so far.Therefore,it is significant to explore the host-virus interaction mechanisms for improvement of disease control.GSK3?,as a highly conserved serine/threonine kinase,is a key regulatory molecule of many cell signaling pathways,and involved in regulating cell differentiation,proliferation,apoptosis,immune response and other physiological functions.In the present study,we cloned the GSK3? homolog from L.vannamei(named LvGSK3?),and analyzed its molecular characteristics and investigated its roles in the process of WSSV infection.The results showed that the infection of WSSV could not only significantly reduce the transcription and translation levels of LvGSK3? in shrimp,but also decrease the phosphorylation levels of LvGSK3?.In order to analyze the role of LvGSK3P in the process of WSSV infection,GSK-3 Inhibitor IX was used as an inhibitor of LvGSK3? activity,and the results showed that the treatment could decrease WSSV infection in the primary shrimp hemocytes.Moreover,the RNAi assays further confirmed that silencing the expression of LvGSK3? could obviously inhibit the the gene expression of WSSV in vivo.In addition,we also found that the treatment would remarkably promote the apoptosis of hemocytes induced by WSSV These results indicated that LvGSK3? might be an important protein molecule for the immunoregulation of shrimp,and shrimp might promote the hemocytes apoptosis to inhibit the WSSV infection by down-regulating LvGSK3?.In conclusion,our study elaborated the function of L.vannamei GSK3? during WSSV infection,which will provide a new clue and theoretical basis for WSSV prevention and treatment.
