Most of insects can be induced to enter diapause by the photoperiod.However,how insects would response to the photoperiodic signal in diapause induction stage remains unclear now.The rice stem borer Chilo suppressalis is an important pest in rice.Larvae of C.suppressalis can induced into a diapause by a short photoperiod and the photosensitive stage is the third instar.In order to probe into the molecular response to photoperiodic signal in diapause induction of C.suppressalis,C.suppressalis larvae in diapause-inducing short photoperiod(LD12:12 h)and nondiapause-inducing long photoperiod(LD16:8 h)conditions were parallelly compared by RNA-seq,q RT-PCR,gene clone.The main results were below:1.The most sensitive stage of C.suppressalis response to photoperiodic signal in diapause induction stageThe switch photoperiod treatment experiments indicated that 9-12 day-old was the most sensitive stage of C.suppressalis to photoperiod at 25 oC.2.The differential expression genes of C.suppressalis response to photoperiodic signal in diapause induction stageTen day-old larvae of C.suppressalis in conditions of LD12:12 h and LD16:8 h photoperiod at 25 oC were used to build comparative transcriptome.Totally,1260 unigene were differential expressed in the C.suppressalis diapause induction stage.Among these differential expressed genes,664 unigene were up-regulated,596 genes were down-regulated in diapause induction condition.From the results,the genes enriched in the Cardiac muscle contraction and Parkinson's disease pathway significantly,the pathway which related with phototransduction like ?ECM-receptor interaction?,?Calcium signaling pathway?,?c GMP-PKG signaling pathway? and ?Phototransduction – fly? were also enriched.The results make contribution to the study of the phototransduction process in C.suppressalis diapause induction stage.3.The clone and expression pattern of the phototransduction related genes in C.suppressalisThe ina D,myosin-IIIb of ?Phototransduction – fly? pathway,PKG of ?c GMP-PKG signaling pathway? and CBP E63-1 were cloned.Their expression pattern indicated that PKG and myosin-IIIb might play a role in the phototransduction in diapause induction stage;CBP E63-1,ina D might play a role in the phototransduction of in post-diapause induction stage.These 4 phototransduction related genes could play an important role in the photoperiod molecular response of C.suppressalis diapause induction stage.In this study,the most sensitive stage of C.suppressalis response to photoperiodic signal in diapause induction stage was determined,differentially expression genes of C.suppressalis response to photoperiod in diapause induction stage were obtained,the expression patterns of phototransduction related genes were analyzed in the C.suppressalis diapause induction stage.These results can contribute to the molecular response to photoperiodic signal in diapause induction stage of C.suppressalis and provide the research material to the study of insect diapause induction molecular regulation.
