| Newcastle Disease(ND)is a highly contagious and acute infectious disease caused by the Newcastle Disease Virus(NDV).NDV can infect a variety of poultry,of which chickens are the most susceptible to NDV.Traditional theory considers that NDV is not easy to infect waterfowl,even if it is infected,the disease does not occur.However,more and more cases of ducks,geese and other waterfowls infected with NDV have been reported in recent years,which has a great impact on the development of the waterfowl aquaculture industry in China.Domestic studies on the effect of duck origin NDV on pathogenicity and immune-related molecules in chickens and ducks are not systematic,and there are some differences between these results.Studies on the effect of NDV on pathogenicity and immune-related molecules in chickens and ducks are conducive to understanding the biological characteristics of duck-derived NDV and the pathogenesis of NDV-infected chickens and ducks.In this study,different virulent duck NDV Duck/CH/HB/JZ-10-1 strains and NDV Duck/CH/HB/JZ-10-2 were used to infect chicks and ducklings,respectively.The chicken spleens were collected at 24 h,48h,and 72 h after infection,respectively.The mRNA transcription levels of 16 kinds of cell immune factors in chickens(IL-1β、IL-2、IL-6、 IL-8、IL-15、IL-16、IL-18、IFN-α、IFN-β、IFN-γ、MHCⅠ、MHCⅡ、STAT1、TLR-7、IRF7、p65)were detected by fluorescent quantitative PCR.Duckling spleens were collected at 1d,3d,and 5d after infection,respectively.The mRNA transcription levels of 9 kinds of cell immune factors in ducks(IL-1β、IL-2、IL-6、IFN-α、IFN-β、IFN-γ、MHCⅠ、MHCⅡ、TLR-7)were detected by fluorescent quantitative PCR..The results showed that when chicks were infected with two strains of duck derived NDV for 24 hours,the mRNA transcription levels of IL-1β、IL-2、IL-8、IL-16、IL-18、IFN-α、IFN-β、MHCⅠ、STAT1、TLR-7 and IRF7 showed a decreasing trend.After 24 hours of infection,the mRNA transcription level increased rapidly,with the highest increase being P65,the mRNA transcription level increased by 235.99 times.Followed by IL-6,after infected with Duck/CH/HB/JZ-10-1 for 24 hours,the transcription level of mRNA increased by 111.88 times,and the mRNA transcription levels of other factors increased by different folds.The detection of mRNA transcription levels of IL-8,IL-15,IFN-γ,and MHCII at different times after infection showed that there was a large difference between the transcription levels at different time points.Further analysis of the data showed that the change trend of mRNA transcription level of different factors after two different virulent virus infection was basically consistent,but there was a certain difference in the amount of increase.After ducklings were infected with two strains of duck derived NDV for 24 hours,the mRNA transcription level of 9 kinds of immune factors(IL-1β、IL-2、IL-6、IFN-α、IFN-β、IFN-γ、MHCⅠ、MHCⅡand TLR-7)declined.After infected for 3d,the mRNA transcription level increased rapidly,with the highest increase being IFN-α,the mRNA transcription level increased by 14.17 times.The mRNA transcription level of other 8 factors showed a different degree of increase from 1 to 14 times.The change trend of mRNA transcription level of different factors after two different virulent virus infection was basically consistent,but there was a certain difference in the amount of increase.After chicks and ducklings were infected with two different virulent duck origin NDV,the mRNA transcription level of chicken immune-related molecules increased more significantly at different time points.Statistic analysis showed that chickens are more susceptible to duck origin NDV infection than ducks.This study will elucidate the changes of immune-related molecules in the host after infected with duck origin NDV and lay a good foundation for the further study of the pathogenesis of NDV infection. |
