Seaweed polysaccharide is extracted from seaweed,it has anti-viral,anti-tumor,anti-clotting effects.In this study,by means of cytokines test of dose screening experiment,contradistinction of the effect of swine fever vaccine,safety tests on target animal(pig)and expanded clinical trials,the impact of seaweed polysaccharide soluble powder on piglets’ immune function and production performance was studied.(1)Experiment of dose screening of seaweed polysaccharide soluble powder involved in immune function of pigBy using different concentrations of seaweed polysaccharide soluble powder(300,200,150,100,50 mg·kg-1,respectively)which were added to diet,the levels of serum interleukins-2(IL-2),interleukin-6(IL-6),interferon-y(IFN-γ),immune globulin,and 1gG were detected to investigate its effect on piglet,.The results showed that on the 7th day,seaweed polysaccharide soluble powder increased significantly the levels of serum IL-2,IL-6,IFN-γ and 1gG,when copmpared with the control group or Astragalus polysaccharide treatment group(P<0.05);Among them,the levels of serum IL-2,IL-6,IFN-γand IgG of the different polysaccharide at dose of 200,150,100mg/kg respectively was significantly increased.On the 14th day,the levels of serum IL-2,IL-6,IFN-y of all treatments with the seaweed polysaccharide soluble powder were significantly increased when compared with the control group(P<0.05);Among them,these treatments at doses of 200,150,100 mg/kg seaweed polysaccharide soluble powder were more obvious when compared with Astragalus polysaccharide treatment,the levels of serum IL-2,IL-6,IFN-y levels of each polysaccharide treatment groups were significantly improved(P<0.05).Compared with Astragalus polysaccharide group,treatment at dose of 150 mg/kg seaweed polysaccharide soluble powder in the diets could increase the serum 1gG levels in animals.By comprehensive analysis of the experimental swine immune index,drug costs etc,we recommend that the dose of seaweed polysaccharide soluble powder in the diets of pig was:100 mg per 1kg,and use for continous 7 days.(2)The effect of seaweed polysaccharide soluble powder on swine fever vaccine inoculationThis experiment was carried out to investigate changes in classical swine fever antibody titer and the levels of serum interleukin-2(IL-2)and interferon-y(IFN-y)in pigs fed feed containing different doses of seaweed polysaccharide soluble powder(200,100,50,0 mg/kg)and Astragalus polysaccharide powder(100 mg/kg).The result showed that the levels of serum cytokines IL-2,IFN-y of the animals treated with seaweed polysaccharides on Day 7,Day 14 and Day 28 were significantly higher than the APS control group,the effect of 100 mg/kg group was best,50mg/kg group was second.It was found that classical swine fever antibody titers of the animals treated with seaweed polysaccharide soluble powder were higher than APS treatmet group or blank control group.The findings showed that seaweed polysaccharide increased the levels of serum cytokines IL-2,IFN-y and classical swine fever antibody titer,when compared with classical swine fever vaccine.The dosage for clinical use of the seaweed polysaccharide as follows:100 mg per each kilometer diet for 7 continous days.(3)Safety test of seaweed polysaccharide soluble powder on target animal(pig)The seaweed polysaccharide at dose of 100,300,500,1000mg/kg were added in swine food to observe whether side affect happened or not,and then blood collection was conducted at the 7th day,14th day.The blood were use for blood analysis and biochemical indices analysis.The result showed that when the diets was added with the seaweed polysaccharide at dose of 100mg or 300mg/kg,the growth performance of piglets was promoted(P<0.05).In the 7th day,the white blood cell count,red blood cell count,hemoglobin and platelet in animals treated with seaweed polysaccharide at 300 mg/kg was higher than other groups(P<0.05).In the 14th day,the white blood cell count,red blood cell count,hemoglobin and platelet of the animals treated with seaweed polysaccharide had no significant difference when compared with each other.Treatment with seaweed polysaccharide at 300mg/kg increased the levels of TP and ALB.The result showed that,by using 1,3,5,10 times of clinical recommended dose for continous 7 days,no toxic signs was observed,and mental condition are fine,appetite are normal.The blood analysis and biochemical indices were in normal range.The findings revealed that treatments with seaweed polysaccharide soluble powder at doses of 1,3,5,10 times of clinical recommended dose had no effect on physiological and biochemical function.By means of comprehensive analysis of the detected indices,seaweed polysaccharide soluble powder was recommended for use at dose of 100 mg/kg diet for use continous 7 days,the safety range of the polysaccharide was broad.(4)A large scale clinical trial of seaweed polysaccharide soluble powder180 of 35~45 day-old pigs were selected,randomly divided to three groups,60 pigs each group.The animals were treated with swine fever vaccine alone at 1.0ml each pig or both seaweed polysaccharide soluble powder at 100 mg/kg diet for use continous 7 days and swine fever vaccinel.Oml for a pig or both Astragalus polysaccharide at 100 mg/kg diet for use continous 7 days and swinefever vaccine at 1.0ml each pig.The result showed that swine fever immune antibody titer in seaweed polysaccharide treatment group was higher than the animals treated with Astragalus polysaccharide.Treatment with seaweed polysaccharide increased the values of swine fever immune antibody when compared with swine fever vaccine alone.
