Probiotics and traditional Chinese medicine extracts have good health care function to the body.It is of profound significance for drug research and development to maintain their respective advantages and application.In this study,the suitable probiotics were selected by comparison test.The probiotics were identified by the conventional method of detecting phenotypic characteristics combined with the modern molecular biology PCR method.The safety of probiotics was evaluated by hemolytic test and mouse safety test.According to the change of the polysaccharide content,the bacterial growth curve and the count of living bacteria count methods,the benefit was observed.The effects of the co culture of the raw bacteria and the traditional Chinese medicine extract were studied,and the effect of probiotics and Chinese medicine extracts on the pathogenic bacteria was studied by the bacteriostasis test.The results were as follows:Test 1.A comparative screening test of 3 probiotics.The probiotics(3 billion cfu/mL)were intragastric administered according to the weight of 0.1mL/10g miceResults there was no significant difference in spleen index and thymus index between the experimental groups(P>0.05).The weight gain of the mixed bacteria group in the seventh day test was 0.72 + 0.04g/d and the weight gain was significantly higher than that of the control group(P<0.05).The weight gain of Clostridium butyrate group was 1.57 + 0.05g/days in Fourteenth days and was significantly higher than that of the control group(P<0.05).In the three stages of Lactobacillus plantarum and mixed bacteria group,abnormal examination of mice appeared.Test two.Identification of Bacillus LYD and resistance test.LYD bacillus was identified as Bacillus licheniformis by traditional methods and molecular biology methods.11 kinds of preservative and disinfectant tolerance test,results 2%sodium hydroxide and 3%hydrogen peroxide had the killing effect on Bacillus licheniformis.In vitro bacteriostasis test was carried out by paper disc method.The results showed that Bacillus licheniformis was not sensitive to amoxicillin and ampicillin in the 24 antimicrobial agents.Haemolysis tests of Bacillus licheniformis were carried out using chicken blood and rabbit blood.A 3 billion cfu/mL concentration of Bacillus licheniformis was given to mice without death.Test three.Co culture of Bacillus licheniformis and astragalus polysaccharides and Forsythia suspense extract.The results of sugar fermentation showed that Bacillus licheniformis could ferment mixed medium containing astragalus polysaccharides and Forsythia suspense extract.MIC test of Bacillus Forsythiae extracts against Bacillus licheniformis showed that the extract of Forsythia suspense at 1mg/mL concentration had no inhibitory effect on Bacillus licheniformis.The inhibition test of the extract of Forsythia Forsythia on pathogenic bacteria showed that it could inhibit the 4 pathogenic bacteria of Staphylococcus aureus,Escherichia coli,Staphylococcus and prowyween.Co culture also inhibited the 4 pathogens.In summary.Screening of LYD Bacillus from three probiotics.The Bacillus LYD was identified as Bacillus licheniformis and could tolerate 9 kinds of conventional disinfectants.Amoxicillin and ampicillin had no antibacterial effect on them,and 3*109cfu/mL of the bacteria were safe in mice.Bacillus licheniformis can be combined with Astragalus Polysaccharide-Forsythia suspense extract.
