The white spot syndrome virus(WSSV)is one of the most vicious viruses causing the devastating death of shrimp.It used to be the vast majority of shrimp culture in Thailand,India,Japan,China,Asia and India and the Pacific.The field caused severe shrimp disease and caused massive deaths of shrimp.The WSSV has a wide range of hosts,mainly infecting many other crustaceans such as shrimp,and even in insects.WSSV has extremely high infectivity and replication ability,and can be widely used to infect subcutaneous tissue,connective tissue,hematopoietic tissue,hemolymph,and antennal glandular tissues of crustaceans such as shrimp.Once infected,WSSV will proliferate and replicate rapidly.From the epidemiological point of view,WSSV transmission can be divided into horizontal transmission and vertical transmission,resulting in a large number of shrimp deaths and huge economic losses that have caused The scientific community has received extensive attention,but the mechanism of invasion of WSSV into host cells remains unclear,severely limiting the development of targeted new antiviral drugs.Therefore,it is important to study the mechanism of WSSV infection in the physiological processes of shrimp and find potential targets for blocking WSSV infection.And influence the survival of infected cells.In this study,crayfish were pretreated with a PI3K-specific inhibitor(LY294002)for comparison to assess the role of the PI3K/Akt signaling pathway in the replication of WSSV.The results showed that on the sixth and tenth days after WSSV infection,the copy number of WSSV in crayfish pretreated with LY294002 was significantly lower than that in Tris-HCl pretreated crayfish.In granulosa cells,the apoptotic rate was increased on the third and sixth days after WSSV infection,and the proportion of apoptotic cells in the LY294002 pretreatment subgroup was significantly decreased on the sixth and tenth days.WSSV infection increased the expression of Bax,Bax inhibitor-1 and lectin mRNA in crayfish blood cells.After secondary stimulation with Tris-HCl,the expression level of Bax in LY294002 pretreated crayfish was significantly higher than that in crayfish pretreated with Tris-HCl on the third or sixth day,but Toll and lectin mRNA were expressed.Volume decreased significantly on the third,sixth and tenth days.On the third day,Bax inhibitor-1 mRNA expression was significantly lower in the LY294002 pretreated group than in the Tris-HCl pretreated lobster group.Together,these results indicate that the host PI3K/Akt signaling pathway plays an active role in balancing WSSV replication between host cell apoptosis and the innate immune response.This information helps to further understand the role of PI3K/Akt signaling pathway in the reproduction of WSSV in crustaceans.
