This experiment was conducted to study the effects of different root cutting treatments on the germination of the lateral root of Paeonia suffruticosa,so as to provide theoretical basis and technical support for lateral root germination of the peony seed taproot.In this research,using Paeonia suffruticosa(‘Fengdanbai')seeds as experimental materials.After the main root germination,root pruning was carried out according to different root length.First of all,select the best way to cut root under the same root length,that is,when the lengt of main root is 2 and 3 cm,making the root cutting treatment with 3/4,1/2,1/3 and 1/5 respectively,while the control treatments were not treated.Then increased in the same way,screening the best root length,namely the main root 1,2,3 and 4 cm long increased 1/3 processing respectively.Determined the key period in the process of lateral root peony seed germination,by morphology and cytology observation in the process of peony seed,and the determination of the peony seed root activity,soluble glycoprotein content,enzyme activity of POD,SOD,PPO,IAAO and the changes of hormones IAA,ABA,GA3 and ZT in the situation,for the research of lateral root peony seed germination rooting mechanism laid a foundation.The main results of test are as follows:1.First of all,through morphological observation,it was found that the root length was 5 cm and the lateral root began to appear,different length of root cutting treatments all formation earlier of lateral root than their respective control,root length of 3 cm root cutting treatment 1/3 lateral root emergence time earlier and the main root is stronger.Through a lot of paraffin section observation found that under the condition of different with lateral root formation are mainly concentrated in 3-5 d,after 3 days root length of 3 cm root cutting treatment 1/3 lateral root primordia formation,and then 5 days later lateral root breaks through the epidermis.2.With peony seeds radicle as experimental materials to research root vigor of germination lateral root of peony seed root by different root cutting treatments.The results showed that the peony roots in the natural sand storage conditions,longer taproot and lower root vigor,when lateral root appeared,root vigor will significantly increased,with the root cutting days increased,compared with the control,the root activity of root pruning increased rapidly at the beginning of lateral root germination,and then decreased,in lateral root germination,root length 3 cm cleared 3/4 root treatment root activity was relatively high.3.According to the change of soluble sugar and protein content,soluble sugar and protein in different root cutting treatments were higher than those of control,and the best way to cut root was root length 3 cm pruning 1/3 root.4.Take the peony seed roots as test materials,changes of enzyme activity during seed germination of peony root of different treatments.The test shows that: the overall change tendency of POD activity most treatments increased first and then decreased,SOD activity changes was "up-down" trend of most treatments,the changes of PPO activity was "up" trend when most processing lateral root germinated,the changes of IAAO activity was "down" trend when most processing lateral root germinated,according to the trend of the enzyme activity,the best way to cut root was root length 3 cm root pruning 1/3 treatment.5.With peony seeds radicle as experimental materials to research endogenous hormones content of germination lateral root of peony seed root by different root cutting treatments.The results showed that with the number of root cutting days increased,the overall trend of endogenous hormones IAA and ZT content is increased first and then decreased,the overall trend of ABA and the content of GA3 was increased firstly and then decreased,according to the trend of the content of endogenous hormones,the best way to cut root was root length 3 cm root pruning 1/3.Comprehensive morphological observation and physiological and biochemical indexes showed that the most beneficial to the peony seed for lateral root germination and growth of way is the best of treatments is 3 cm root cleared 1/3 treatment.
