A total of 32 normal-weight Arbor Acres broilers(AA broiler,average weight:1.86kg)and 46-day-old silky fowls(abbreviated as silky chickens,average body weight: 0.75 kg)were selected,and randomly assigned to 6 treatment groups: AA broiler control group(PBS injection,n=10),AA broiler low-dose insulin treatment group(80 ?g/kg body weight,n=10),AA broiler high-dose insulin treatment group(100 ?g/kg body weight,n=6),silky chicken control group(PBS injection,n=12),silky low-dose insulin treatment group(80 ?g/kg body weight,n=12)and silky high-dose insulin treatment group(100 ?g/kg body weight,n=12).In order to study the effect of body weight on blood glucose levels,serum biochemical parameters,and gene expression in poultry,low-weight AA broiler(average body weight: 0.88 kg)were selected,and low-dose insulin(80 ?g/kg body weight,n=6)was designated as low body weight group.Starvation treatment was performed for 12 h at the age of 46 days,and insulin resistance tests were performed at 47 days of age.The test is divided into the following two parts:(4)the effect of different insulin doses on the relative gene expression in AA broiler and silky.The test is divided into the following two parts:Trial 1 Effects of Different Insulin Dose on Blood Glucose Level and Serum Biochemical Index of AA broiler Chicken and silkyIn order to study:(1)the effect of pancreas index on blood glucose regulation in AA broiler and silky(2)the effect of different insulin doses on blood glucose in AA broiler and silky,(3)the effect of different insulin doses on serum biochemical markers in AA broiler and silky.The test results are as follows:(1)Pancreatic index: The low-body recombinant pancreas index of AA broiler was significantly higher than that of the normal body AA broiler,which was about 1.72 times that of normal body AA broiler.AA AA broiler cantaining low-body was no significant difference in pancrea index compared with silky.The pancreas index of silky is significantly higher than that AA AA broiler of normal weight,which is about 1.31 times that of the normal weight group.There is no significant difference between the AA AA broiler of low body and silky in pancrea index.(2)The blood glucose content: At 120 minute the AA broiler and silky were fed,the blood glucose level of AA broiler remained unchanged.The blood sugar content of black chickens increased significantly.When 240 min,the blood glucose content of silky was significantly higher than that of AA broiler(P<0.01),which is about 2.56 times of that of AA broiler.The influence of body weight difference on blood glucose content: in the natural state,the blood glucose of high body weight was significantly(P<0.05)higher than that low body weight group.In the AA broiler and silky of the same weight level,the normal blood glucose level of silky was significantly higher(P<0.01)than that of AA broiler.(3)The Serum biochemical indicators: the UREA in Serum of Insulin high-dose group in silky was significantly higher(P<0.05)than that of AA broiler,which was about 3.57 times than that of AA broiler;the Insulin in Serum of Insulin high-dose group in AA broiler was significantly higher(P<0.05)than AA broiler of Insulin low-dose;the UREA in the serum was significantly higher in the silky group(P<0.01)than in the AA broiler group both at 120 min and 240 min,and was about 3.54 and 2.65 times that of the AA broiler group.at the minute of 240 the UREA in serum of silky was significantly higher(P<0.05)than that in AA broiler.at the minute of 240 the URIC in serum of silky was significantly higher(P<0.05)than that of 0min,which was about 2.09 times that of 0 min.At the minute of 120,the CHOL in serum of silky was extremely significantly higher(P<0.01)than that of AA broiler,which was about 1.48 times that of AA broiler.In silky,the CHOL in serum of 120 min was significantly higher(P<0.01)than that at240 min,which was about 1.58 times that of 240.(4)Effect of weight difference on serum biochemical indicators: at the minute of 240 The GLU in serum of the low body AA broiler was significantly higher(P<0.05)than that of the high body AA broiler,which was about 1.84 times that of the high body AA broiler.at the minute of 240 The UREA in serum of silky was high body AA broiler extremely significantly higher(P<0.01)than that of silky,which was about 2.93 times that of low body AA broiler,The URIC in serum of silky was high body AA broiler significantly higher(P<0.05)than that of silky,which was about 1.99 times that of low body AA broiler.Trial 2 Effects of Different Insulin Dose on Gene Expression in AA broiler and silkyIn order to investigate the effect of insulin dose on gene expression of AA broiler and silky,at the age of 47-day-old the experiment collected liver,hypothalamus,pectoral muscle and pancreas of healthy AA broiler and silky at the minute of 0min,120 min and 240 min.Primers were designed based on the related genes(GLUT2,GLUT12,NPY,and Insulin Receptor)of the poultry's insulin resistance,and quantitative analysis of candidate genes and related sequences was performed by using fluorescent quantitative RT-PCR.The test results are as follows:The relative expression of GLUT2 in the hypothalamus of AA broiler low dose group was significantly higher(P<0.05)than that in the high dose group,which was about 11.51 times that of the high dose group.In the low dose group,the relative expression of GLUT2 of AA broiler was significantly higher(P<0.05)than silky.Which is about 11.68 times that of silky.The relative expression of NPY in the hypothalamus of Insulin low-dose silky was significantly higher(P<0.05)than that of the high-dose group,Which is about 3.78 times that of the high-dose group.The relative expression of GLUT2 in the hypothalamus of AA broiler at 240 min was extremely significantly(P<0.01),higher than that of 120 min,and significantly(P<0.05)higher than that of 0 min,which was about 7.48 times that of 120 min and 3.06 times of that 0 min;At the minute of 240,The relative expression of GLUT2 in the hypothalamus of AA broiler was significantly higher(P<0.05)than that of silky,which was about 11.67 times that of silky.Effect of Weight Difference on Gene Expression in AA broiler and silky: At the minute of 240,the relative expression of GLUT2 in the hypothalamus of high body weight AA broiler was significantly higher(P<0.05)than that in the low body weight,which was about 8.28 times that of the low body,whereas the relative expression level of IN in the low body weigh was significantly higher(P<0.05).P<0.05)than that of high body weight group,which was about 2.13 times that of high body weight group.In the pectoral muscle,the relative expression of the low body recombination Insulin Receptor was significantly higher(P<0.05)than that of the high body weight group,which was about 2.56 times that of the high body weight group.
