As a very important cut flower,Lilium is widely cultivated around the world.However,the Lilium is a flower that is susceptible to diseases,and it is often affected by various pathogens in production and cultivation.The most serious and common disease is the bulb rot,which will lead to the rot of lily bulb.With increasing interest about lily,there are more and more researches concerning lily.The whole industry has developed profoundly.Because of the influence of disease,the development of lily has been hindered.So it is vital to cultivate resistant varieties in China.In this paper,we used different pollination experiments of interspecific hybridization in different hybrid varieties.In accordance with the result,we observed the viability of pollen,studied pollination method and improved the way of hybrid embryo rescue so as to overcome the intergroup incompatibility.And distant hybridization lily as the research object,we have carried out the biological characteristics,disease resistance and other related research.The main contents and results were as follows:1.By morphological identification and ITS sequence analysis,it can be confirmed that the pathogen of Lilium bulb rot from the institute of agricultural science of Zhuzhou is Fusariμm oxysporum.2.The results of comparison of seven different inoculation methods showed that the inoculation method of stabbing and soaking seeding was more suitable for the identification of resistance to bulb rot of lily.The results of identification of resistance to bulb rot of 60 Lilium hybrids showed that LO hybrids(Lilium Longiflorum × Lilium Oriental)were the most resistant to the diseases;It is true that the disease resistance of OT(Lilium Oriental × Lilium Trumpet)hybrids and edible lilies was generally low,a few was at the level of moderate resistance,and most were at the level of susceptibility.3.According to the result,the cut-ovary pollination method has the advantage of overcoming incongruity before fertilization and get the swelling capsules easier compared to other three methods which included general pollination,cut-chapiter and chapiter graft method.The post-fertilization gap and the compatibility of the parents could affect the interspecific cross too.4.The distant hybridization test of L×O(Lilium Longiflorum × Lilium Oriental)hybrid combination(L.Snow Queen × L.Marco Polo and L.Snow Queen × L.Siberia,L.Snow Queen × L.Sorbonne)was successful,The methods of in vitro pollination,embryo rescue and tissue culture were used in the study,the hybrid seedlings were cultivated after the three procedures of bourgeon,proliferation and rooting by the embryo rescue the utilization of the tissue culture in order.5.Cytological observation of F1 hybrids produced from OT×O showed that the aneuploid plants were obtained and its chromosome number varied from 16~48 from Manissa×Siberia,and the triploid plants were obtained and its chromosome number is 2n = 3x = 36 from Robina×Sorbonne.6.In the majority of asexually propagated progenies from OT×O,their mitosis were regular,but some abnormal phenomena,such as unequal division,chromosome bridges,lagging chromosomes,nucleus germination,intruding and splitting of the tube-like nucleus and multiple nuclei were observed.These abnormal phenomena during mitosis may be one of the mechanisms of producing chromosome number and structure variations among the asexual descendants derived from OT×O.
