Arachidonic acid?ARA?is a precursor of many active substances and plays a very important role in living organisms.The rice field eel is an important economic animal of special aquatic products in China.It has been farmed on a large scale,the more the food is produced,the faster it grows,and the faster it dies,the more likely it is to develop"fatty liver"or"hepatobiliary syndrome"disease.In the previous study,the research group found that ARA has a certain liver protection effect on rice field eel,to understand the effect of ARA on the liver protection and its mechanismof rice field eel,and also provides the basis for the rational preparation of compound feed of the rice field eel,The research results from four aspects are as follows.1.The effects of different proportions of ARA on the performance of the rice field eel were added.The proportion of ARA was 0?control group?,0.35%,0.70%,1.05%,1.40%and 1.75%of the combined feed,feeding the young eel with 30d.Results:ARA had significant influence on the growth and survival rate of the young eel,and the comprehensive analysis showed that the appropriate proportion was 0.70%.2.The effect of 0.70%ARA on growth and physiological and biochemical indexes of rice field eel was added.In the experiment group,0.7%ARA was added to the basal feed,and no ARA was added to the control group.The growth of rice field eel,muscle nutrition,liver function and lipid metabolism were measured.Results:the addition of ARA can promote the growth of rice field eel,reduces muscle moisture content,and increases muscle protein and fat content;decrease the liver body index and visceral index;increases the activity of glutamate transaminase and glutamate transaminase in liver tissues;lower serum total cholesterol and increase HDL.It is shown that adding ARA can enhance liver function and enhance fat metabolism,reduces protein as energy consumption and promote protein synthesis,thus improving the growth performance of rice field eel.3.The effect of ARA on liver injury of rice field eel.Through which was fed with ARA in feed,was used as experimental group,and those without ARA as control group,inject CCl4 liver injury agent in the same time.observed the changes of liver microstructure and measured the serum biochemical indexes and observed the expression of related genes in the metabolic pathway of ARA in the rice field eel over time.The results showed microstructure shows that experimental liver cell integrity and cell membrane structure more complete than the control group,experimental group liver cell damage ratio significantly less than control group,and cell damage degree is low,and structure of the liver tissue repair time is short.The activity of GPT and SOD in the serum of the experimental group was higher than that in the control group.MDA content was lower than the control group.PLA2,COX-1 and COX-2 were significantly higher in 6,12and 24h after the injury,and no significant difference was found between the normal level and the control group of 48h.5-Lox,12-Lox and CYP1a were significantly higher than the control group.Show that ARA mechanism of eel protecting liver,lies in the maintenance of liver cell membrane integrity,improve liver immunity,especially through the COX-1,COX-2 metabolic pathways by raising SOD activity and reduce MDA content,reduce the oxidative damage of cell,promote discharge poisonous and harmful material At the same time,through 5-LOX,12-LOX and CYP1a pathway,it can promote the discharge of toxic and harmful substances,accelerate the repair rate of damaged cells,and restore the liver function more quickly.4.Effects of adding earthworm on the growth performance of rice field eel in feed.In order to understand the application of earthworm containing high proportion of ARA in body fat as a source of ARA.It was fed with 0%,25%,50%,75%and 100%of earthworm for 61 days.Results:compared with the control group,the weight gain rate and specific growth rate of 25%earthworm in the feed group were significantly different.The maximum fertilizer content of the feed group with 50%earthworm was significant.According to the regression results of growth and fertilizer abundance,35%earthworm can obtain the best production performance.
