| In order to better prevent and treat the liver disease in the process of rice field eel(Monopterus albus)in the breeding process.In this paper,the histology,chemical reagent and fluorescence quantitative PCR technology were used to studied theliver development process of rice field eel,liver injured and effects of PLA2,COX1,COX2,5-LOX and CYP1 A gene expression on the metabolism of Arachidonic acid in liver tissue by carbon tetrachloride.The results are as follows:(1)The developmental characteristics of the liver in Monopterus albus.Results showed that consistent with most bony fishes,Liver of rice field eel started to develop in the newly hatched young rice field eel before,after gradually perfect.The right side of the digestive tract can be observed to stain the deeper layers of liver cells at day 1 after hatching young rice field eel.You can see hepatic sinusoid at day 3 after hatching young rice field eel.Liver begin to store the posterior part of the liver lipid at day 5 after hatching young rice field eel.Liver accumulation of fat is more obvious at day 7 after hatching young rice field eel.The liver close to the development of yolk sac,with the development of rice field eel liver,yolk sac gradually disappear and showing a close contact.The liver is well developed,has been generally mature.The basic structure of the liver has been formed when when the yolk sac disappeared.3 months after hatching young rice field eel.During the breeding period,the size of the liver cells of the maturely ovum female rice field eel was larger than that of the immaturely ovum female rice field eel.There was no significant difference in the microstructure of hepatic cell of the female rice flied eel at the same year of non-breeding period.(2)Effects of different CCl4 concentrations on Microstructure of liver tissues on Monopterus albus:The acute degeneration of hepatocytes induced by CCl4 in experimental rice field eel showed a pathological change characterized by typical vacuolar degeneration,nuclear deviations,disappearance of nucleolus and incomplete nuclear membrane,and increased with the increase of concentration.(3)The effects of different carbon tetrachloride concentrations on the serum and liver tissue biochemical indexes were examined in Monopterus albus:The experimental group serum index compared with the control group,body mass and body mass of 0.254g/kg 0.508g/kg carbon tetrachloride concentrations6-48 h solution injection can cause serum GPT,GOT,CAT,LDH activity increased and MDA content;the 6-96 h activity of SOD decreased,and GPT,GOT,CAT activity and MDA content reached the maximum at 12h(P<0.01),LDH reached the maximum value.SOD reaches the minimum at 24h(P<0.01).Compared with the control group,body mass and body mass of 0.254g/kg 0.508g/kg carbon tetrachloride concentrations 6-24 h solution injection can cause the liver LDH activity and MDA content increased significantly(P < 0.01);GPT,GOT,CAT and SOD activity decreased significantly,and reached the peak at 12(P<0.01)h,the liver index in 12 h raised or lowered the maximum value multiple(P<0.01).Then slowly returned to the control group with no significant difference.(4)Effects of different CCl4 concentrations on the expression of PLA2,COX-1,COX-2,5-LOX and CYP1 A genes of Arachidonic acid metabolism of Monopterus albus:Using gene cloning and study of fluorescence quantitative PCR showed that injection of carbon tetrachloride solution 6h PLA2 gene on liver of rice field ell expression had no obvious change,COX-1,COX-2,5-LOX and CYP1 A gene expression was inhibited(P < 0.01);12hPLA2,COX-1,COX-2,5-LOX and CYP1 A gene expression was induced by high concentration of low concentration of 24hPLA2,COX-2 and inhibitory state;the expression of 5-LOX gene was induced by high concentration of low concentration to inhibit the expression of 12 h,but is more close to the control group,COX-1 group and CYP1 ABecause the expression were inhibited;48-144 h,low concentration group the expression of PLA2 gene induced by 96h(P < 0.05),PLA2,expression of COX-1,5-LOX and CYP1 A genes were inhibited;288hPLA2,COX-1,COX-2,5-LOX and CYP1 A gene expression compared with the control group had no obvious change. |
