Ceroplastes japonicus Green(Hemiptera,Coccidae,Ceroplastes Gray1828)parasitics on a variety of plants,and has a wide range of damage.C.japonicus sucks nutrient from plant phloem sap which is not enough to meet the growth of C.japonicus.The endosymbionts of C.japonicus can supplement these nutrient-poor substances.This paper aims to discuss the diversity of endosymbionts communities in the C.japonicus and their effects on the host nutrient metabolism.In order to provide a theoretical basis for the biological control technology with endosymbionts as the target.In this experiment,high-throughput sequencing technology was used to analyze the diversity of the endosymbionts by sequencing the variable region of the 16 S rDNA.The material for the diversity research of endosymbionts in different hosts of C.japonicus are the nymphs from the persimmon tree and the jujube tree respectively.The material for the diversity research of endosymbionts in different ages of C.japonicus are the nymphs and the adults from the persimmon tree.In order to analyze the effects of endosymbionts on the nutrient metabolism to its host,the nutrient changes in the haemolymph of C.japonicus before or after antibiotics treatment were detected.The results as follows:1.The diversity of C.japonicus' endosymbionts from different hosts.The dominant phylum of the C.japonicus from the persimmon tree were Bacteroidetes and Proteobacteria,and the dominant phylum of the C.japonicus from the jujube tree were Bacteroides and Firmicutes.After antibiotic treatment,the abundance of Bacteroidetes decreased from 41.2% to29.6%,and the abundance of Betaproteobacteria decreased from 17.5% to12.16%.The 16 S function prediction shows that Proteobacteria,Bacteroidetes,and Firmicutes may be related to host drug resistance.Bacteroides may participate in the metabolic pathways of the nymphs of the jujube tree,and Candidatus Uzinura may be related to the synthesis of aminoacids in the nymphs of the jujube tree.Chlamydiales may take part in specific biological processes.Alpha diversity analysis shows that the nymphs of the jujube tree was higher in species diversity than the persimmon tree.After treatment with antibiotics,its species abundance was reduced.The Beta diversity analysis shows that the effect of antibiotic treatment on the nymphs of jujube trees was greater than that of persimmon trees.2.The diversity of C.japonicus' endosymbionts from different ages.In adult stage,the dominant phylum of C.japonicus were Firmicutes,Bacteroidetes,and Proteobacteria which were identical to the nymph.After the antibiotic treatment,?-Proteobacteria abundance of adult stage decreased from 23.88% to 6.93%,and the abundance of Burkholderia dropped from22.35% to 3.62%.The 16 S function prediction shows that Rhizobiales may participate in the host's nitrogen fixation,Bacillus may have the ability to degrade cellulose,Rickettsia may increase host's heat resistance,and Bdellovibrio may have bactericidal action.The treatment with antibiotics had greater effect on adults than the nymphs.3.Effect of endosymbionts community changes on host nutrient metabolism.After treatment with antibiotics,the concentrations of glucose and albumin in the hemolymph of C.japonicus decreased significantly,and the triglyceride concentration increased.In this experiment,the dominant bacteria,population structure,and the prediction of bacterial function in different hosts,different ages and antibiotic treatment of C.japonicus were obtained.It was verified that the endosymbionts affected the nutrient metabolism of C.japonicus.It was established different hosts of C.japonicus have the best age of prevention.
