In order to understand the genetic structure among the three different populations of Hu sheep and carry out the next conservation work,the diversity of the three Huyang populations in Anhui Province was tested using RAPD molecular marker technology,the genetic diversity among populations and individual differences within groups was analyzed.According to the existing literature references,25 polymorphic random primers were selected.During the experiment,10 reproducible bands with clear polymorphic primers were selected for RAPD analysis,A total of 96 Hu sheep genomic DNAs were amplified in 3 populations.According to the Shannon diversity index,population genetic diversity was calculated,and Nei's formula was used to calculate the genetic distance index between varieties.Specific strong fragments were selected for sequencing and BLAST analysis was performed.Ten primers produce polymorphic markers of varying degrees.A total of 61 DNA amplification fragments were obtained from these 10 primers,and at least 4 fragments were amplified from each primer,with a maximum of 8 fragments.On average,6 DNA fragments were amplified from each primer.The fragment length ranged from 500 to 2000 bp.The total number of polymorphic loci in the entire population was 52,accounting for 85.25% of the total loci.Among them,47 polymorphic sites in Zhoushi Hu sheep farm,accounting for 77.05%;44 polymorphic sites in Yushan Hu sheep farm and accounted for 72.13%;47 polymorphic sites in Yunxing Hu sheep farm accounted for 77.05%.Conclusions:(1)In Zhoushi Hu sheep farm,The Nei's gene diversity index was 0.2949 and the Shannon information index was 0.4340.In Yunxing Hu sheep farm,The Nei's gene diversity index was 0.2941 and the Shannon information index was 0.4348.In Yushan Hu sheep farm,The Nei's gene diversity index was 0.2815 and the Shannon information index was 0.4135.The diversity index between the two bodies in the population was compared,and the order of the genetic diversity was Zhoushi>Yunxing>Yushan.(2)The genetic similarity index between the Zhoushi Hu sheep farm and Yunxing Hu sheep farm was 0.9832 and the genetic distance was 0.0169.The genetic similarity index between the Zhoushi Hu sheep farm and the Yushan Hu sheep farm was 0.9631;The genetic distance was 0.0379.The genetic similarity index between Yunxing Hu sheep farm and Yushan Hu sheep farm was 0.9658 and the genetic distance was 0.0348.(3)There are variations in the similarity of the three populations,of which the Zhoushi sheep farm has a greater similarity to the Yunxing sheep farm.(4)The sequences of the 43 u chromosomes 1,3,8,12,17,23,and 24 isolated from the OVIS of bighorn sheep have high similarity to chromosomes 5,16,17 and 24 isolated from Tibetan yak.
