To study the effect of glucose oxidase on yellow feather broilers,1-day-old 900 healthy yellow feather broilers were selected and randomly divided into 3 groups with 5 replicates of 60 chickens each.The test period was from 1 to 63 days.These 3 groups in the experiment were control group,virginiamycin group and glucose oxidase group.The control group was fed with corn pod-based diets and 400 g/t virginiamycin and 300 g/t glucose oxidase(1000 U/g enzyme activity)were added to the basal diet separately.The growth performance,slaughter performance,and immune organ index of broilers test were recorded,and the intestinal tract morphology,cecal chyme microflora,tight junction proteins,and cytokine mRNA expression were measured.The effect whether glucose oxidase could replace virginiamycin in yellow feather broilers' growth performance were studied from intestinal microflora,immune function and so on.The test results were as followed:1.Glucose oxidase increased the average daily gain and daily feed intake,and reduced F/G in the late period(43~63 days)compared with the control group and the virginiamycin group.The difference was not significant(P>0.05);2.The glucose oxidase group had higher semi-paralytic rate,breast muscle ratio,and leg muscle ratio than the control group and virginiamycin group.The slaughter rate and net nevus rate were higher than the control group but lower than the virginiamycin.Tthe difference was not significant(P>0.05);3.The bursal index and spleen index of glucose oxidase group were higher than virginiamycin group but lower than control group,and the difference was not significant(P>0.05);4.The pH in glandular stomach of glucose oxidase group was lower than control group and virginiamycin group(P>0.05).While the pH in the duodenum,jejunum,ileum and cecum of the glucose oxidase group was significantly higher than virginiamycin group(P<0.05).The pH of the ileum was significantly higher than the control group(P<0.05);5.The villus height and VH/CD in glucose oxidase group was higher than control group and lower than virginiamycin group.The crypt depth was higher than the other two groups.The villus height in the virginiamycin group was significantly higher than control group(P<0.05);6.The OTU value of glucose oxidase group was significantly higher than control group and virginiamycin group(P<0.05);the relative abundance of Bacteroidetes was significantly lower than control group and virginiamycin group(P<0.05)and the ratio of Firmicutes and Bacteroidetes was increased(P>0.05);glucose oxidase reduced the relative abundances of Bacteroidaceae and Porphyromonadaceae,while the relative abundances of Rikenellaceae,Ruminococcaceae,Lachnospiraceae and Desulfovibrionaceae were lower than control group but higher than virginiamycin group;the relative abundances of Prevotellaceae and Synergistaceae were lower than virginiamycin group and higher than control group(P>0.05);the relative abundances of Veillonellaceae in virginiamycin group was significantly higher than control group and glucose oxidase group(P>0.05);7.The relative expression of ZO-1 mRNA in duodenum and ileum was higher in glucose oxidase group than control group and virginiamycin group,and that in jejunum was higher than control group but lower than virginiamycin group.The relative expression of occludin mRNA in small intestine of glucose oxidase group was significantly higher than control group,and that in duodenum and jejunum were significantly higher than virginiamycin group(P<0.05).The relative expression of occludin mRNA in duodenum were significantly lower in virginiamycin group than control group but higher in jejunum(P<0.05);8.The relative expression of IL-1? mRNA in small intestinal of the glucose oxidase group was lower than the control group and that in ileum was lower than virginiamycin group;the relative expression of IL-4 mRNA in the duodenum was lower than control group and higher than virginiamycin group;IL-4 in jejunum and ileum and intestinal IFN-? mRNA were higher than control group and virginiamycin group;IL-8 in duodenum was lower than control group,while the relative expression of IL-8 mRNA in jejunum was higher than control group(P>0.05).In summary,adding 300 g/t glucose oxidase to diets had effects on the cecal chyme microflora,intestinal tight junction proteins,and intestinal immune factors to improve intestinal tract morphology,so as to improve performance and can replace virginiamycin to some extent.
