Petunia(Petunia hybrida)is a very beautiful ornamental plant.It is often used as a bed plant during important holidays.Tethering factor plays an important role in the vesicle transportation.VPS52 is member of GARP(golgi-associated retrograde protein)complex family.COG8 is member of COG(conserved oligomeric golgi)complex family.They play an important role in the tethered process.However,little is known about the role of VPS52 and COG8 in species other than Arabidopsis.In this work,we invested the function of PhVPS52 in petunia,a homolog of the Arabidopsis VPS52 gene,and explored the establishment of the genetic transformation system of petunia.The PCR assay of plants showed that the PhCOG8 had been integrated into petunia and nicotiana(Nicotiana tabacum).The results are as follows:(1)PhVPS52 silencing influenced the growth and development of petunia.Specifically,the bases of stems were brownish,the leaves were yellow,and there was irregular deep purple stripes on the petals.PhVPS52 silencing resulted in abnormal development of chloroplasts.(2)Genetic transformation system of petunia 'Mitchell' was established.The concentration of kanamycin of leaf differentiation and screening was 50 mg/L.The optimal concentration of rooting culture was 60 mg/L.The concentration of cefotaxime sodium salt was 500 mg/L and the 1-naphthylacetic acid concentration in rooting culture was 0.4 mg/L.(3)The insertion sites of exogenous genes can be found by Tail-PCR detection.We obtained some transgenic petunia and nicotiana.
