Pharmacokinetics Of Cefquinome/Sulbactam Combined In Swine

In this study,the pharmacokinetics of cefquinome and cefquinome/sulbactam in swine were compared.Liquid chromatography tandem mass spectrometry?HPLC-MS/MS?Methods were developed for the determination of cefquinome and sulbactam in porcine plasma.The chromatograms of cefquinome and sulbactam are of good peak shape with less interference from peripherals,which meets the need for determination of drugs in plasma.The correlation coefficients were greater than 0.999 in the range of 1⁵00 ng/mL.The limits of detection?LOD?for cefquinome and sulbactam were 0.1 ng/mL and 0.25 ng/m and the limis of quantitation?LOQ?were for that 0.4 ng/mL and 0.5 ng/mL.The extraction recovery of cefquinome in plasma was 81.92%⁹8.91%,the intra-assay coefficient of variation was 0.98%⁵.16%,and the inter-assay coefficient of variation was 4.99%⁶.47%.The extraction recovery rate of sulbactam in plasma was from 81.30%¹10.24%,the intra-assay coefficient of variation was 0.93%⁴.93%,and the inter-assay coefficient of variation was 2.96%⁷.60%.Six healthy pigs were randomly divided into two groups and were received 2 mg/kg b.w.of cefquinome sulfate suspension and 2 mg/kg b.w.cefquinome sulfate suspension/10mg/kg b.w.sulbactam sodium by intramuscular injection and Cross-administration for the pharmacokinetic test.The main pharmacokinetic parameters of cefquinome in healthy pigs after intramuscular injection of cefquinome sulfate suspension and Combined injection of cefquinome sulfate suspension/sulbactam sodium were:t1/2?were 2.79±0.68 and 2.91±0.24 h,T_max were 0.67±0.13 and 0.37±0.13 h,C_max were 2.19±0.17 and 2.72±0.36?g/mL,AUC_0-last were 7.02±0.40 and 7.39±0.91?g·h/mL,CL/F were 0.28±0.01 and0.25±0.03 L/h/kg,Vd/F were 0.76±0.30 and 0.83±0.24 L/kg,MRT were 3.14±0.37and 3.16±0.09 h.The main pharmacokinetic parameters of sulbactam after combined intramuscular cefquinome sulfate suspension/sulbactam sodium were as follow:t1/2?=3.55±0.51 h,T_max=0.38±0.14 h,C_max=18.76±2.01?g/mL,AUC_0-last=2.05±5.14?g·h/mL,CL/F=0.32±0.05 L/h/kg,Vd/F=1.65±0.45 L/kg,MRT=1.65±0.09 h.The results showed that cefquinoxine was absorbed rapidly,peaked shortly and eliminated quickly in pigs after single and combined intramuscular injection.The main pharmacokinetic parameters of cefquinome in pigs such as t1/2?,T_max,C_max,AUC_0-last,CL/F,Vd/F,MRT were not statistically significant?p>0.05?after single and combined intramuscular injection.The half-life of sulbactam and cefquinome was almost the same after the combination therapy.This study shows that the combined use of cefquinome and sulbactam does not affect the pharmacokinetics of cefquinome in pigs and is suitable for combination therapy.
