The brachionus plicatilis cultivated with baker's yeast lack the highly unsaturated fatty acids that is needed by sea fish larvae,which is easy to cause a low survival rate of brood fish.Meanwhile,most of the artemia nauplius lack the unsaturated fatty acids.This text researches the influence of the larval Hybrid Giant Tigers' survival rate,body composition,digestive enzyme and fatty acid by feeding concentrated chlorellas,cod-liver oil and capsules that contain deep-sea fish oil.It also studies how above three enhancers have an effect on the body composition and fatty acid of brachionus and artemia,including discuss the time and dose to enhance the artemia.It aims to filtrate suitable enhancer to improve the fish larvae's survival rate,and calculate the certain time and dose for enhancer to provide theoretical basis for production.The experiment is divided into three parts,whose main content is as follows:1.By using the analytical method,the strengthened brachionus and artemia'body composition is measured.Simultaneously,the analysis for brachionus and artemia'fatty acid is measured by using gas chromatography.Strengthening time and dose of artenia is discussed,too.The results show that the water content of brachionus and artemia in the control group is higher than the test group(P<0.05),three enhancers make inapparent difference in crude protein's content(P>0.05).The group that fed with cod-liver oil and capsules that contain deep-sea fish oil contains more crude fat than the control group and concentrated chlorellas group(P<0.05).However,the enhancers have a remarkable difference in brachionus' s saturated fatty acid(P<0.05).And it's marshalling sequence is as follows:group that with capsules that contain deep-sea fish oil>concentrated chlorellas group>cod-liver oil group>control group.The brachionus in enhanced group has a higher monounsaturated fatty acid content than the control group(P<0.05),it's marshalling sequence is as follows:group that with capsules that contain deep-sea fish oil=concentrated chlorellas group>cod-liver oil group>control group.The enhanced group contains more polyunsaturated fatty acids content than the control group(P<0.05),the marshalling sequence is:group that with capsules that contain deep-sea fish oil > cod-liver oil group=concentrated chlorellas group > control group.Three enhancers make unconspicuousc differences to artemia's saturated fatty acid(P>0.05),the artemia enhanced with cod-liver oil has a lower monounsaturated fatty acid content than the other groups(P<0.05)while the three enhanced groups have more polyunsaturated fatty acids than the control group obviously(P<0.05),it's marshalling sequence is:group that with capsules that contain deep-sea fish oil>concentrated chlorellas group>cod-liver oil group.The time when the artemia enhanced with concentrated chlorellas makes no significant effect on it's saturated fatty acid and monounsaturated fatty acid(P>0.05).But if enhanced for more than six hours,it contains more polyunsaturated fatty acids than the control group(P<0.05).The time when the artemia enhanced with capsules that contain deep-sea fish oil makes unconspicuous differences for it's saturated fatty acid and monounsaturated fatty acid(P>0.05)while it has apparent differences when enhanced more than twelve hours(P<0.05).Similarly,the time that cod-liver oil takes to enhance artemia has the same result as the former.The research results manifest that three enhancers can reduce the water content of brachionus and artemia,but cod-liver oil and capsules that contain deep-sea fish oil can increase it's crude fat content.Besides,the enhancers can't affect the crude protein of the brachionus and artemia.Three enhancers can improve the brachionus and artemia's polyunsaturated fatty acid content,however,they can't improve the content of saturated fatty acid apparently.They even have no linear effect on monounsaturated fatty acid.Moreover,the enhancing time of concentrated chlorellas should be longer than six hours while capsules that contain deep-sea fish oil and cod-liver oil should be longer than twelve hours.2.Adopting the conventional measurement method by measuring body composition,the Hybrid Giant Tigers' body composition are measured.And by applying gas chromatography method,the fatty acids of fish is analyzed.Besides,the body length,weight and survival rate is counted.The result shows that fish larvae in the enhanced group have a higher survival ratio than the control group(P<0.05),but it's fish larvae contain fewer water than the test group(P<0.05).There are apparent differences among the groups(P>0.05),yet concentrated chlorellas group's crude protein is apparently higher than the other groups(P<0.05).The test group's survival rate is significantly higher than the control group(P<0.05),and it's body length is longer and it's weight is heavier than the test group(P<0.05).In addition,the condition factor among the groups is indistinctive(P>0.05).The saturated fatty acid and polyunsaturated fatty acids of the groups is indistinctive(P>0.05).Oppositely,the group with capsules that contain deep-sea fish oil and cod-liver oil group contain more saturated fatty acid and polyunsaturated fatty acids than the control group and the concentrated chlorellas group(P<0.05).The study results indicate that enhancers can improve the Hybrid Giant Tigers' survival rate,body length and weight,but make no influences for it's fatness,saturated fatty acid and polyunsaturated fatty acids.3.Applying conventional measurement method,Hybrid Giant Tigers' digestive enzyme is measured and the influences on fish larvae's digestive enzyme by feeding with three enhancers are analyzed.Meanwhile,some digestive enzyme are investigated conditionally.The result displays that the entero-amylase of each group has inapparent differences(P>0.05),the concentrated chlorellas group' s erepsin and pepsin are more active than the other groups(P<0.05).The group fed with capsules that contain deep-sea fish oil and cod-liver oil group have more lipase activities than the control group and concentrated chlorellas group(P < 0.05).When the temperature is 40?,it has more pepsin,erepsin,entero-amylase and lipase activities than the other groups(P<0.05).When the concentration of substrate of pepsin is 0.5%,it's activity is more active than the other groups in different temperature(P<0.05).When the concentration of substrate of entero-amylase is 0.5%,it has more activities than the other groups in different temperature(P<0.05).If the p H of pepsin is 2.6,it has more activities than the other groups in different temperature(P<0.05).If the PH of erepsin is between 9.35 and 12.32,it is more active than other groups(P<0.05)while entero-amylase's p H is 8.0,it is more active than other groups(P<0.05).If lipase's p H is 7.4,it is more active than other groups(P<0.05).The research results show that above three enhancers can increase the survival rate of Hybrid Giant Tigers,hel P the fish grow longer and add more weight.However,the enhancers make no influences to fish's saturated fatty acid and polyunsaturated fatty acids,and they have no linear influences on monounsaturated fatty acid.Three enhancers have no influences on lipase's activity.The concentrated chlorellas can improve the activities of pepsin and erepsin,capsules that contain deep-sea fish oil and cod-liver oil can increase the lipase's activities.
