A total of 200 healthy 1 week old pheasants(females)were randomly divided into 5 groups.They were bred for 16 weeks at the experimental base of Yan'an University.The feed amount of five groups of feed was 0%,4%,8%,12% and 16% respectively.During the experiment,free drinking water was added,and the feed was added on time.After the feeding test,Each group randomly selected 12 pheasants,After 12 hours of fasting and weigh,The carotid artery bloodletting to death,Quickly remove the required tissue materials.The effects of mega grass feed on growth index and fat related gene expression of pheasant were studied by paraffin section and real-time fluorescence quantitative PCR.It provides a reference for the allocation of Pennisetum giganteum feed.Test 1 Effect of Pennisetum giganteum feed on the growth performance of pheasant(1)growth properties: The effects of Pennisetum giganteum feed on daily gain and average weight of pheasant were significant,but not on average body weight and material weight ratio.(2)Physiological and biochemical determination:Physiological measurement,the influence of Pennisetum giganteum feed on WBC? HCT?MCHC?MPV ?MCV and PDW were obviously affected by pheasant,no significant effect on RBC and RDW-SD.Biochemical determination,the effect of Pennisetum giganteum feed on TP?ALB?GLO?Urea?TG?TC?HDL-C and LDL-C were significant by pheasant,and the effect on GLU was not significant.(3)Determination of immune organs: Influence of Pennisetum giganteum feed on spleen and thymus tissue were obviously by pheasant,but no significant effect on Bursa,kidney and liver tissue.(4)Determination of intestinal villi: Each was Paraffin sections through observation duodenum,jejunum and ileum used for pheasant.The effect of Pennisetum giganteum feed on villus height of jejunum,duodenal and ileum of pheasant were significan;The villus width of duodenum,jejunum and ileum was not significant;The effect of villus crypt depth on duodenum and jejunum was significant,But there was no significant effect on villus recess depth in ileum.Test 2 Effect of Pennisetum giganteum feed on H-FABP gene,A-FABP gene,LPL gene and CAST gene expression of pheasantTest animals and test one,Using rt-pcr method to study Pennisetum giganteum feed of H-FABP,A-FABP,LPL and CAST gene expression level in different organizations.The results are as follows:Study found that H-FABP,A-FABP,LPL and CAST gene in pheasant stomach muscle,myocardial,chest and leg muscle were expressed.A-FABP gene,The highest expression in the myocardium,stomach muscle and chest muscle tissue were found in 8% Pennisetum giganteum feed,the highest expression in the leg muscles tissue was found in the 12% Pennisetum giganteum feed,but the increase was significant in the breast muscle tissue;H-FABP gene,The highest expression in the stomach muscles and chest muscles tissue were found in 8% Pennisetum giganteum feed,the highest expression in the myocardium and leg muscles tissue were found in the 12% Pennisetum giganteum feed,but increased significantly in the myocardium,stomach muscles and the leg muscles tissue.LPL gene,The highest expression in themyocardium and leg muscle tissue were found in 8% Pennisetum giganteum feed,the highest expression in the stomach muscles and chest muscles tissue were found in the 12% Pennisetum giganteum feed,but increased significantly in the stomach muscle and chest muscles tissue.CAST gene,The highest expression in stomach muscle tissue was found in 8% Pennisetum giganteum feed,the highest expression in the myocardium,chest muscle and leg muscle tissue were found in the 12% Pennisetum giganteum feed,but it was significantly increased in myocardium,stomach muscle and leg muscle tissue.
