| The experiment was conducted to study the Effects of Serotonin on Bone Calcium Mobilization of Ewe in Transition period,and furthermore,to study influence mechanism of organic selenium on milk performance and to provide theoretical basis and technical guidance for scientific applications of serotonin in practice of sheep feeding.Under the present experimental conditions,the results showed as follows:The experiment one was used the ANKOM gas production system in the United States to identify nutrient composition of the diet,the rumen fermentation parameters in vitro,and the CNCPS(Connell net protein and carbohydrate system in the United States)were used to dissection the proportion of the internal nutrient structure in the diet,and the total gas yield was added to the total yield by Logistic-dual pool model.The other experiment,under the above experimental diet,was conducted using a single factor randomized arrangement,and thirty Bamei ewes at the same transition period(14 days before birth)to be randomly divided into 3 groups with 10 replicates in each group.On the 7d before delivery to 0d day,the 5-HTP group intraveous 0.178mg/kg BW,the TRP group also intraveous 0.178mg/kg BW,the control group added the normal saline.Both 5-HTP and TRP were injected into the jugular vein of sheep.The bjective of this study was to explore the effects of 5-serotonin on the performance of ewes in transition period,the indexes of calcium transport in the breast and femur,and the expression of bone calcium mobilization related genes.Under the present experimental conditions,the results showed as follows:(1)The average daily gain of lambs was significantly increased,and the milk yield of ewes was also increased after 5-HTP perfusion.(2)The concentration of calcium and 5-HT in blood and milk and PTHrP concentration in blood can be significantly increased after perfusion with 5-HTP and TRP.(3)Compared with the control group,the 5-HTP group significantly increased the mRNA expression of the gene PTHrP,NCX1,PMCA2,SPCA1,CaSR and the bone reabsorption marker related gene RANK-L in the mammary gland of the ewes,and the gene expression of PTHrP,PMCA2,SPCA1,SPCA1,etc.in the TRP group also increased significantly. |
