In order to explore the influence of environmental p H conditions on gene expression of Suillus luteus,sequenced the transcriptome of strain growed in different p H environment.And the result is that,compared with the hyphae grown in p H 3.0,the hyphae at p H 4.0,4.5,5.0,5.5,6.0,7.0 and 8.0 had 743,870,1177,1512,1628,3135 and 2902 genes were differentially expressed respectively,.These differential expression genes were respectively annotated to 2029,2368,2733,3125,3168,4626 and 4728 functions in gene ontology,and set to 76,81,108,121,116,180 and 177 metabolic pathways in the Kyoto Genome and Genome Encyclopedia.Based on the differential expression of its genes,we infer mycelial changes in the activity of enzymes,mechanisms of induction environmental p H,and differences in physiological metabolism.The results showed that the content of glyoxylate was consistent with the oxalic acid production,and presented positive correlation between the two based on the changes in the expression levels of the gene encoding ICL and MS.Higher levels of glyoxylate,succinate,oxaloacetate and malate help the mycelium which are at p H 6.0,7.0,and 8.0 to adapt and regulate the p H of the environment by the expression changes of the gene encoding FUM,CS,ICL and MDH.The MAPK signaling pathway,chloride channel,ABC transporters,G protein-coupled signal transduction and MFS substrate transporter on the plasma membrane of mycelia are activated to sense the p H of the environment.Fatty acid metabolism,porphyrin and chlorophyll metabolism,steroid biosynthesis,taurine and hypotaurine metabolism and butanoate metabolism,pyruvate metabolism,glycine,serine and threonine metabolism,and many metabolic pathways had high activity at p H 4.5 and 6.0,that were important factors for optimal growth of mycelium under p H 4.5 and 6.0 conditions.
