Osmanthus fragrans?Thunb.?Lour,belonging to O leaceae,is one of the traditional top 10 flowers in Chinaand have been grown widely for landscape in Guangdong province where is located on the south-subtropical zone.However,less systematic research on Osmanthus fragrans in this province have been reported up to date.Therefore,It is significant to initiate a survey and study onthe germplasm and application of Osmanthus fragransin Guangdongfor enriching the researcheson Osmanthus fragrans researches,exploring potential variety resources,as well as facilitating the application of Osmanthus fragrans in this region.The dissertation generally investigates cultivars resources in Guangdong with prefecture-level citiesas the sample center.The application in Guangdong landscape of Osmanthus fragranswere analyze based on the statistics of grown cultivar group,percentage used in different type of green spaces and so on.The trait of main components and genetic relationships were analysed by the way of trait coding.The DNA polymorphism and genetic relationships of 60Osmanthus fragransmaterialswere analysed usingscreened10 ISSR marker primers.The results were shown asfollows:1.A long history could be dated back to the cultivation of Osmanthus fragrans in Guangdong,which leaded to abundant resources of local species.The distribution of cultivation was distinguished by the tropic of cancer.Thecultivars resources of Osmanthus fragranswas relatively less in south of the tropic of cancer while species amount and growing density increased with the latitude increasing northward.Studying on cultivars resources of,77 trees of Osmanthus fragranswere recorded and collected which of 27belonged toOsmanthus fragransAsiaticus Group,25 inOsmanthus fragrans Letus Group,18 in Osmanthus fragrans Albus Group,and 7 in Osmanthus fragrans Aurantiacus Group.2.The application of Osmanthus fragrans in Guangdong was in superiority of Osmanthus fragransAsiaticus Group with ratio above 70%in most regions.TheOsmanthus fragransin south of the tropic of cancer apperaed in low frequency.There was only 64.2%of Osmanthus fragrans in the west of Guangdong where Osmanthus fragrans Albus Group and Osmanthus fragrans Aurantiacus Group wereoddly planted.The utilization ratio increased in accordance with the latitude increasing northward.Eastern region of Guangdong even shared a ratio up to 91.4%where Osmanthus fragrans Letus Group,Osmanthus fragrans Albus Group,and Osmanthus fragrans Aurantiacus Groupwere significantly multiplied.The arranged planting contributed 40%,and the opposite planting contributed 21.4%of the planting form.3.56 of collected 63 samples could be figured out based on the trait analysis and cluster analysis.16cultivarswereidentifiedfromthose materials.NY-S-01?S?wasmostsimilarto Osmanthusfragrans‘Danzhuang'.GM-S-01,SG-S-01,Osmanthus fragrans‘Zi Yingui',and Osmanthus fragrans‘Changye Bizhu'had the most discrepancy in trait with othe rs.Osmanthus fragrans‘Danzhuang'and NY-S-02,NY-S-03?S?and NY-S-03,as well as NY-S-04?S?and N Y-S-04 were most likely male and female plant in the same cultivar.The kinship between Osmanthus fragrans Asiaticus Group and autumnal Osmanthus Group was far.On the other hand,in the group of autumn Osmanthus,The kinship Osmanthus fragrans Albus Group and Osmanthus fragrans Letus Group was close.4.10 of DNA polymorphism primerswerescreened by ISSR?Inter Simple Sequence Repeat?marker for Osmanthus fragrans.Observed number of alleles,effective number of alleles,Nei's genediversity parameter,Shannonn diversity parameter,and percentage of polymorphic loci order of 4 species groups were:Osmanthus fragrans Letus Group>OsmanthusfragransAlbusGroup>OsmanthusfragransAsiaticus Group>Osmanthus fragrans Aurantiacus Group.53 of collected 60 samplescould be figured out based on ISSRmarker.The genetic similarity coefficient between Osmanthus fragrans‘Danzhuang'and NY-S-04 was 0.95,which could identify they werethe same cultivar.Osmanthus fragrans‘Danzhuang'and SG-S-05,NY-S-04?S?and Osmanthus fragrans‘Yuegui',as well as II03–S-05 and NY-S-01 were all sharing a close genetic similarity coefficient of 0.9,which might be the same cultivar.Thegenetic similarity coefficient and cluster analysis demonstrated that the result of ISSR marker was similar to that of trait quantity classification.
