To investigate the effects of Sansui duck 100 kDa transmembrane glycoprotein receptor gene(PMEL-17),dopamine receptor gene(DRD4)and serotonin type 1A receptor gene(5-HT1A)in the tissues of Sansui ducks.To further explore the mechanism of occurrence of Sansui duck behavior.This research applied molecular genetic methods and applied SPSS and Excel and other bioinformatics analysis software to the sansui duck PMEL-17,DRD4 and 5-HT1 A genes in the sansui duck heart,liver,spleen,lung,kidney,intestine,brain.The biological analysis was performed on the expression of 12 tissues in the chest,legs,eyes,muscle stomach and glandular stomach.Sixty-six ducks were selected for observation of behavior and normal behavior of slaughtered feathers.Three slaughtered ducks were selected,after slaughter(one plume behavior and two normal behaviors)and their tissues were collected.RNA was used to perform RNAi on these tissues using Trizol method.After extraction,a cDNA library of Sanshou duck was constructed,and the expression levels of the above three genes in related tissues were detected by fluorescent quantitative PCR.Finally,the complete coding region(CDS)of PMEL-17 gene of Sansui duck was cloned by RT-PCR and vector construction method.The results were analyzed by bioinformatics.The following results are obtained:1.The 5-HT1 A receptor gene expression was highest in the pupal behavioral brain tissue and the expression difference was extremely significant(P<0.01),followed by renal tissue,and the expression results were significantly different(P<0.05).In the remaining 10 tissues,the expression of the gene was similar and the5-HT1 A gene had no significant difference compared with the normal behavior group(P>0.05).2.The expression of DRD4 receptor gene was highest in the pupal behavioral brain tissue and the expression difference was extremely significant(P<0.01).The expression of this gene in the pupal behavioral group was much higher than that in thenormal behavioral group.In the remaining 11 tissues,the expression level of the gene was similar in the normal behavior group and the abnormal behavior group,and there was no significant difference in the DRD4 gene expression compared with the normal behavior group(P>0.05).3.PMEL-17 receptor gene expression was highest in the pupal behavioral brain tissue and the expression difference was extremely significant(P<0.01),and the expression level of the PMEL-17 receptor gene in the pupal behavioral group was much higher than normal behavior.group.In the remaining 11 tissues,the expression level of this gene was similar in the normal behavior group and the abnormal behavior group,and there was no significant difference in the expression of PMEL-17 gene compared with the normal behavior group(P>0.05).4.There are two synonymous mutation sites in the CDS region of the PMEL-17 gene of Sansui Duck.The secondary structure of the SSR duck has changed slightly,but the structure of the translated protein has not changed..In summary,the abnormal behaviors of the 5-HT1 A,DRD4,and PMEL-17 receptor genes on the sansui ducks.There may be significant regulation,and changes in the structure of the CDS domain of the PMEL-17 receptor gene may not have an effect on the abnormal behavior of the Sansui duck.
