Amelanchier sinica is a deciduous tree belonging to the apple subfamily of Rosaceae,one of only two species of Amelanchier plants produced in China It is native to Henan,Gansu,Shaanxi,Hubei and Sichuan.The plant is 3-15 meters high and grows on slopes and shrubs at an altitude of 1000-2000 meters.Its leaves are ovate,inflorescences are racemose,its flower heads are pendulous,its petals are slender,white and aromatic,and the blooms in May,with good ornamental value.Fruit is near spherical or oblate,fruit period is about September to October,the calcium content of the fruit is very high,and contain multivitamin,trace element and sugar lipoprotein,have high economic value.the study aimed to establish and optimize a tissue culture system for Amelanchier sinica.In the a large number of seedlings breeding can be carried out.The plant germplasm resources of Amelanchier sinica are protected and utilized,and the species of tree species in China can be enriched.Taking Amelanchier sinica embryo tissue of somatic embryo callus(embryoid)as test materials,using the method of in vitro culture,to study the influence of different disinfection methods,basal media,phytohormone on shoot proliferation and rooting,and different time of opening bottles,substrate compositions,rooting time on transplanting of tissue cultured seedlings of Amelanchier asiatica.The experimental results are as follows:1.The best disinfection method was 75% Ethanol for 30 s and 0.1% HgCl2 for 8 min,the germination rate reached 17.3%.2.The best breeding medium for sterile test tube was: MS + 0.5mg/L 6-BA + 0.02mg/L NAA + 0.05mg/L IBA + 35g/L sucrose + 7.5g/L agar;the proliferation coefficient reached 6.28 and the seedling growth was robust,the stem was thick and strong,the leaves were large and bright green,without yellow leaves and vitrification.3.The most suitable medium for sterile test tube seedlings was 1/4MS + 0.5mg/L IBA + 30g/L sucrose + 7g/L agar;the rooting rate reached 100%,the rooting average length reached 3.5cm,the main root was robust,and there were fibrous roots.4.Choose healthy growth,after rooting culture 25 d,high in 3 cm above the test-tube seedlings and bottle to 24 h after seedling transplanting,the most suitable ratio of matrix volume of refining plants are as follows: domestic peat: domestic water moss: humus = 1:5:1,survival rate reached 70%.
