Grape(Vitis vinifera L.)is one of the momentous economic fruit trees and diffusely planted all over the world.Especially after it has been confirmed that resveratrol in grape berries is the first of all kinds of fruits and the important role of resveratrol in the prevention and treatment of cardiovascular disease and cancer,it has promoted the market consumption of grape and related products.In the international market,seedless grape has become the popular direction in industrial development and breeding research.At present,the outstanding problem is that the seedless grape varieties are single,especially lack of high quality,large grain and red seedless grape.Therefore,it is vast significant in the study of the formation mechanism of seedless grapes and identification of the genes which related to seedlessness.This study was based on the transcriptome of the key period of ovule development of the seeded and seedless grape hybrids,screening and cloning a gene VvYABBY4 which expresses differences,its function has been preliminary verified.The chief results in this study are discreibed as followings:1.According to the whole genome of grape,we identified 7 YABBY genes and divided them into 5 subfamilies based on phylogenetic analysis as well as the former research.Genes in the same subgroup have similar protein motifs and exon/intron structures.Addition,it is conclude that several grape YABBY genes arose before divergence of the two species based on the analyse of synteny between the Arabidapsis and grape genomes.Due to the expression analysis of YABBY genes in the period of ovule development in 4 cultivars,VvYABBY4 genes had a significant expression between seeded and seedless varieties in the critical period of ovule abortion.Furthermore,there were also varied expression in different organs,which suggest YABBY genes play diverse roles in the development of multiple tissues in the grape.2.VvYABBY4 was cloned in seed grape Red Globe and seedless grape Thompson Seedless.The ORF of VvYABBY4 was 552 and encode 183 amino acids.Ecotopic expression of VvYABBBY4 in tomato(Micro-Tom)showed some different phenotypes compare with wild ones: Transgenic plant became dwarf,apical dominance went into weaken,the leaves got more dark green,the serration of the edge of leaves goes weakening.Compare with the control,the inflorescence enlargement,however,vitality of pollen and pollen germination capacity had no significant differences.Addition,the fruit reduced slightly and seeds to be smaller in size distinctly.According to the Real time quantitative PCR,expression patterns of tomato genes which associated with seed development were found varied in transgenetic tomato lines compared with control,suggesting VvYABBY4 gene may affect the expression of related gene to co-regulate seed development.
