| Contagiouts ecthyma(ce or orf),is an exanthematous zoonotic disease caused by the orf virus.In the disease,the pimples,blisters,and pustules are formed around the lips and nose of the diseased animal.Finally,verrucous scabs are formed,and proliferative lesions are produced in the hairless parts of the nose and mouth.The lambs are impaired more easily due to their own immune system is unfit.After infected by orfv,it is difficult for lambs to eat,to fully intake of nutrients,seriously affecting the growth and development of lambs,and ultimately the body lacks nutrients,starving to death.Repeated infections and secondary infections of contagious pleuropneumonia,conjunctivitis,etc.may occur in the same host,and secondary infections increase mortality several times.The ORF virus is also detectable in the blood of asymptomatic goats,indicating that asymptomatic animals can also be carriers of the ORF virus.Laoshan dairy goats,one of the fine varieties of dairy goats in China,have been bred in large quantities in Shandong’s Jiaodong peninsula.In recent years,ORF have occurred in a number of farms in the area,causing huge economic losses to goat breeding.However,it is not known the epidemic status of ORF in this area.Therefore,this study firstly used the goats in the Otter Goats Farm of Qingdao as a sample to conduct a molecular etiological investigation of ORFV,and divided 200 apparently healthy adult pregnant sheep into nonimmune ones 100 as control groups and the other 100 as immune test groups.Anticoagulated blood samples were collected before and after immunization.DNA was extracted from the samples.The laboratory established PCR method was used to detect the pathogens of the ORFV.Meanwhile,laboratory-separated FX0910 ORF virus were used to culture and amplificate on bovine testicular cells and inactivated with a final concentration of 0.4% formaldehyde solution;Cells were inoculated with inactivated venom to detect the inactivation effect of the virus;Preparate of inactivated vaccine with the adjuvant of transfer factor and aluminum glue salt mixture.Safety of inactivated vaccine was tested by injecting with 5 times immunization doses to healthy goats.Pregnant goats in the experimental group were immunized twice,and as the control group,anticoagulation was collected about 14 days after the first immunity and the second immunity.Blood was used to detect the pathogen of ORF virus;serum of the two groups was separated and the neutralizing antibody titer was detected;During the parturition season,cord blood of two groups of newborn lambs was collected to detect the pathogen of the ORF virus and the protective effect of vaccine immunity was determined to the lamb.The results of this study are as follows:1.The detection rate of the ORF virus in the peripheral blood was 21% in the peripheral blood samples before immunization,and 4% in the peripheral blood samples after the second immunization;the detection rates of the ORF virus before and after the immunization were 17%,18%;2.The neutralizing antibody titers of the inactivated vaccine before immunization,after the first immunization and after the second immunization were 1: 1.89,1: 4.92,and 1: 27.86,respectively;3.The detection rate of umbilical cord blood was 0 in the immunized group and 9.7% in the unimmunized group.In conclusion1.Through this study,we have ascertained the status of the pathogens of ORF virus in goats farms,and detected the pathogen detection rate of pregnant goats without clinical symptoms;2.Inactivated vaccine of transfer factor and aluminum colloidal salt adjuvant of ORF virus was prepared,and the inactivated vaccine was safe and reliable through experiments;3.Vaccination of pregnant goats with inactivated vaccine of ORF virus can significantly increase the level of neutralizing antibodies in pregnant goats,and at the same time,it can reduce the rate of infection of goats,indicating that the vaccine has a certain pathogen removal and treatment of ORF virus;4.The detection rate of the newborn lambs in the immunized group was significantly lower than that in the non-immunized group,indicating that the prepared inactivated vaccine against ORF had a certain degree of immune protective effect on lambs. |
