Matrine alkaloid is water-soluble total alkaloids isolated from Sophora such as Sophora flavescens Ait.and Sophora alopecuroids L.,including matrine,oxymatrine,sophocarpine and sophoridine and so on.It has a wide range of medical and agricultural activities.In its pest control spectrum,its activity against aphids is outstanding,however the mechanism is not clear yet.Thus,we taked Acyrthosiphon pisum as tested insect with matrine,sophocarpine and cytisine as tested agents to determine the effects of three alkaloids on the neuro-enzymes of Acyrthosiphon pisum to provide basis for the research of insecticidal mechanism of matrine.The results were shown as follows:?1?Matrine and sophocarpine showed similar poisoning symptoms:convulsions,paralysis,death and significant symptoms of poisoning appeared after 20 minutes.The symptoms of poisoning caused by cytisine were similar to that of matrine,but stronger convulsions.?2?Three kinds of alkaloids showed some inhibitory effects on AchE.In vivo,showed significant inhibitory effects and time effect,but the inhibition rate was less than 35%.In vitro,matrine and sophocarpine had no obvious inhibitory effect on AchE,and the inhibitory effect of cytisine at 1 mg/mL concentration was significant?34.05%?.?3?Three kinds of alkaloids showed no obvious effects on the content of Ach.?4?Three kinds of alkaloids showed some inhibitory effects on Na+,K+-ATPase.In vivo,showed significant inhibitory effects and time effect and inhibition intensity:cytisine>sophocarpine>matrine.The inhibition rates under different treatment time periods were14.86%,29.29%,and 33.81%?matrine?,22.58%,29.51%,and 36.74%?sophocarpine?,20.50%,31.20%,and 46.83%?cytisine?respectively.In vitro,they showed concentration-dependent.Cytisine showed strongest inhibition,0.01,0.1 and 1 mg/mL?the same below?inhibitory rate was 25.56%,45.59%and 62.73%,respectively.Matrine Second,the inhibition rates were 16.42%,35.99%,and 48.41%,respectively.The inhibition of sophocarpine was the weakest,inhibition rates were 17.20%,31.86%,and 39.29%,respectively.?5?Three kinds of alkaloids showed some inhibitory effects on Ca2+,Mg2+-ATPase.In vivo,showed significant inhibitory effects and time effect and inhibition intensity:cytisine>matrine>sophocarpine.The inhibition rates under different treatment time periods were17.61%,25.49%and 33.81%?matrine?,14.46%,22.06%and 30.60%?sophocarpine?,20.04%,38.76%and 43.22%?cytisine?,respectively.In vitro,showed significant inhibitory effects and concentration-dependent and the inhibition rates were 22.72%,33.70%and 50.87%;22.52%,31.28%and 44.59%;22.52%,26.96%and 47.11%,respectively.?6?Three kinds of alkaloids showed some inhibitory effects on ALT.In vivo,showed significant inhibitory effects and time effect and the inhibition rates were 19.31%,30.03%,38.40%?matrine?,17.04%,26.31%and 38.77%?sophocarpine?,23.90%,26.87%,and 23.35%?cytisine?,respectively,but cytisine showed less than 30%.In vitro,matrine and sophocarpine showed significant inhibitory effects and concentration-dependent,the inhibition rates were26.21%,38.22%,50.74%and 27.27%,37.40%,57.34%,respectively.After treated with cytisine,as the concentration increased,it showed a certain inhibitory effect,and the inhibition rates were 11.58%,20.85%,and 37.16%,respectively.?7?After treated 10 h,the activity of GAD was not significantly different from that of control.Afterwards,the activity of GAD increased,the specific activity was 1.22 and 1.11times?matrine?,1.20 and 1.18 times?sophocarpine?,1.14 and 1.20 times?cytisine?higher than that of the control.?8?Three alkaloids caused a significant increase in Glu content,which was a 0.5 to 1time increase compared to the control.Afterwards,the increase decreased,the specific activity was 0.33 to 0.67 times higher than that of the control.?9?After treated 10 h,three alkaloids caused a significant increase in GABA content,a 1to 1.3 time.However,the content of GABA after treatment with cytisine was little different with control for 20 h.When treated 30 h,matrine and sophocarpine had little difference with control and the content of GABA was reduced by cytisine.These results showed that the toxicity effect of matrine co?Ld relate to its effects on the nervous system and the GABA and Glu systems may be the main site of action and Na+,K+-ATPase and Ca2+,Mg2+-ATPase may also be one of its targets.Sophocarpine is similar to matrine,and cytisine is generally similar to matrine,and there are differences in individual index.
