This study selected Valsa mali Miyabe et Yamada as tested fungi,screened biological activity of 20 kinds of plant alcohol extracts and futher studied Kochia scoparia(L.)Schrad,such as the separation of active ingredients and mechanism of exploration.The results indicated:1.The inhibition rate of 20 kinds of plant ethanol extracts against to Valsa mali Miyabe et Yamada is filtered by the mycelium growth rate method.The result showed that:15 species of plant extracts showed inhibitory effect on the mycelia growth;the inhibitory effect of Kochia scoparia extract was most obvious,antibacterial rate were 92%,under the concentration of 2.00mg·mL-1.2.The antifungal activities of the extract from Kochia scoparia against Valsa mali Miyabe et Yamada was tested.The result showed that:the petroleum ether extract of Kochia scoparia against Valsa mali Miyabe et Yamada was 96.5%,the chloroform extract of Kochia scoparia against Valsa mali Miyabe et Yamada was 75.2%and the Methanol extract of Kochia scoparia against Valsa mali Miyabe et Yamada was 94.3%.The minimal inhibitory concentration(MIC)of this three extracts were 68.7mg·L-1,137.6mg·L-1 and 95.1mg·L-1.3.The silicagel column was used to separate and purify active constitutes of the extraction of the petroleum ether extract.A total of 8 fractions were seen by TLC analysis.The bioassay showed that the flow L7 have strong antifungal activities,EC50 of which was 1.1mg·L-1.4.Treated by flow L7 with a concentration of 0.1 mg·mL-1,as shown by biological microscope,the mycelium of tested fungi were irregular with deformity and inclusions leaked.Treated by flow L7 with different concentrations,hyphae cell membrane permeability increased,at the same time,the content of soluble proteins,pyruvic acid and reducing sugar in mycelia were reduced in different degree.
