Ethanamizuril is a new triazine anticoccidial drug independently developed by the Shanghai Veterinary Research Institute,Chinese Academy of Agricultural Sciences.EZL is a high-performence,low toxic,safe,broad spectrum anticoccidial drug and has a broad research prospects,according to the preliminary pharmacological toxicological studies.In order to provide more data support for clinical rational drug use and new veterinary drug declaration of EZL.The UPLC-UV methods were developed for the detection of EZL in plasma of beagle dog and EZL,M3 in plasma of chicken in this paper to analyze the pharmacokinetic characterristics of EZL in beagle dog after oral administration EZL of low,medium and high doses.Furthermore,the effects of three different dispersants on the exposure of EZL in beagle dogs and chickens,and the regulation of concentration changes of EZL and M3 in chicken plasma at different feed additive dose were studied.In this paper,beagle dogs were oral administrated with three doses of 0.3,0.6,3mg/kg bw of EZL and the pharmacokinetices were analyzed by the UPLC-UV method.The results showed that the drug peak time(Tmax)was 1 h,the half-life(t1/2)was 2 h,and the maximum plasma concentration(Cmax)and the area under the drug-time curve(AUC0-t)had a good linear relationship with the oral dose.There was no significant difference between the male and female beagle dogs on pharmacokinetics.The oral dose of beagle dogs and chickens was 3 mg/kg and 1.33 mg/kg respectively to research the exposure of three different dispersants?water,0.5%CMCNa,PEG200?.The results showed that Tmaxax in beagle dogs and chickens was 1 h,and 4-5h respectively.t1/2/2 was 2-3 h and 9-11 h respectively.AUC(0-t)related to drug exposure had no statistical difference?P>0.05?both in beagle dogs and chickens,indicating that the dispersants?water,0.5%CMCNa and PEG200?used in the study did not have statistical difference on effect of EZL exposure in beagle dogs and chickens.In this study,the UPLC-UV method were developed to detect EZL,M3 in the chicken plasma and researched the regulation of concentration changes of EZL and M3in chicken plasma at different feed additive doses.The results showed that the concentration of EZL and M3 at the feed additive dose range of 3-200 mg/kg was in a good linear relationship as the feed addtive dose increased,the linear relationship between growth trend of EZL and M3 was good and basically maintained synchronous growth.
